46 diploid tartary buckwheat accessions(Fagopyrum tataricum) were used in chromosome doubling experiment. The differences of morphology, cytogenetics, seed protein content, seed flavonoid content between tetraploid and corresponding diploid accessions were studied. The main results were as follows:1.12 new accessions of tetraploid tartary buckwheat were created successfully and identified. They are all autotetraploid,2n=4x=32.2.Morphological comparison showed that there are significant difference of 1000-seed weight between autotetraploid and diploid tartary buckwheat, but no significant differences of seed number per a plant, seed weight per a plant, branch number in main stem, and sub-branch number.3.The results of cytology and pollen fertility showed that the rate of abnormal tetrads and the average micronuclei number per tetrad in tetraploid tartary buckwheat are all much higher than those in diploid tartary buckwheat. The chromosome pairing configuration at PMC MI of autotetraploid tartary buckwheat is generally 4x = 8â…£.The rates of defeated pollen grains in tetraploid tartary buckwheat are all much higher than those in diploid tartary buckwheat.4.Kjeldahl analysis showed that the seed protein contents of tetraploid tartary buckwheat are much significantly higher than the diploid. The seed protein contents of tetraploid tartary buckwheat ranged from 15.1%to 19.4%,with an average of 17.21%.And the seed protein contents of diploid tartary buckwheat have the range from 12.9%to 15.9%,with an average of 13.97%.5.Seed flavonoid content analysis showed that the seed flavonoid contents of tetraploid tartary buckwheat are much significantly higher than that of the diploid tartary buckwheat. The seed flavonoid contents of tetraploid tartary buckwheat ranged from 3.12%to 4.57%,with an average of 3.64%,and those of diploid buckwheat are from 2.42%to 2.98%,with an average of 2.77%.
