| The babesiosis is a tick-borne zoonosis blood protozoiasis.The bovine babesiosis is reportedly the one of the severest parasitosis that does harm to the world cattle industry. 1980, Levy and Ristic established micro-gas-phase static culture,but also achieve the continuous culture in vitro Babesia parasites and has been the desired results. After the method was quickly applied to Babesia bigemina, Babesia canis,Babesia equi,etc. have been successfully cultured in vitro. At present,the method researchers used babesiosis separation and detection of the pathogen,the immune diagnosis,immune surveillance,antigen component analysis,drug screening,helminth preservation.Therefore,the technology known as Babesia research milestone in the history. Xinjiang,local Babesia bovis has not yet been able to cultivate in vitro studies, this study was try to explore the methods and conditions of the cultivation of Xinjiang babesia bovis in vitro. And make a preliminary investigation to epidemiology.The following three studies were conducted in the present article.1. Karamay region investigation of piroplasmosis.There are three types of ticks spread of bovine piroplasmosis:Ixodes persulcatus, Hyalomma asiaticum and Dermacentor nuttalli. This disease mostly occrus mixed infection,seasonal clearly. Incidence of the season from the beginning of May,reached a peak in mid-June,after mid-July non-disease occrus. in September once the peak,then there is no disesae infection.2 .The PCR Detection of babesia bovis.In this study,through the PCR method detected bovine Babesia,Babesia bigimina,Theileria annulata infected mixture.We selected the singleness infection sample for study of the cultivation in vitro.3 .The Cultivation of babesia bovis in vitro.The results were shown that cultivation method of babesia bovis in vitro could be established by conditiongs of the fresh blood which contained babesia bovis,complete medium which contained 40% bovine serum and cultivation at 37℃in 5%CO2.The cultivating babesia bovis could be survived for 14 days and subcultured for 13 times.The maximum percentage of parasitemia was reached to 12.5% and average value of parasitemia was 4%.This experiment on the cultivation of babesia bovis in vitro studied several outside factors affecting the comparison,the results show that: the pH value of the culture medium significantly affected the growth and propagation of babesia bovis,the optimum pH for the in vitro is 7.0~7.2;fitting serum concentration is 40%;inactivated serum Babesia supporting the growth of capacity is low than the non-inactivated serum;The effect of cultivation is the same between physical solidification of the serum and the natural solidification of the serum;There are no significant difference at the supporting Babesia growth capacity of the culture medium of RPMI-1640 and M-199. |
