Laser is a different natural light radiation. And Laser irradiation is a traditional method for crop improvement. Intrinsic DNA methylamine pattern is an integral component of the epigenetic network in many eukaryotes. Study on variation of DNA methylation pattern will help us a comprehensive understanding of function of DNA methylation on the apparent biological control. Here, we used molecular technology to study the possible both genetic and epigenetic alterations induced by laser irradiation.In this paper, Oryza. Sativa japonica JL01-124 has been treated by laser irradiation with different wave length and time. Several phenotypic mutants appeared in the treated plants, such as dispersed-spikelets, earlier-ripening and elongated kernel, etc.We also found that both genetic variation and DNA methylation patterns were altered in some of the laser-irradiated rice M0 plants. Genome-wide fingerprinting on methylation patterns by the MSAP method revealed that the methylation alterations were actually widespread, and also mainly represents demethylation.Based on the above observations, we conclude that laser irradiation represents an effective mutagen for rice, not only inducing classical sequences alterations, but also the epigenetic changes, such as, alterations in DNA methylamine patterns. Therefore, the study may provide some reference information for designing irradiation-based crop mutational breeding.
