This paper determined the content of rotenone in different parts of Amorpha frutiicosa L by Reversed-Phase High Performance Liquid Chromatography.Studied on qualitative and quantitat ive methods and extracting procedure. By the determined way,researchde on rotenone distribution and variation during different periods and different parts of Amorpha frutiicosa L and the toxicity of extract on Larvae.And further researched the compatibility and the activity of defensive enzymes in Larvae.Results showed:The optimum conditions were as follows,extacting temperature was 40℃, extracting time was 30min,extacting solvent was chloroform,supersonic wave frequency was 100kHz.The process was simple, reliable and high determination.It was reason able technique;In RT-HPLC,the YWGC18(250mm×4.6mm,id,10μm)columnand methanol-acetonitrile-0.03mol/L phosphoric acid solution(30:32:38) as mobile phase were used.The flow rate was 1.0mL/min and the detective wavelength was 295nm.The column temperature was 25℃. Under the chromatogra phic conditions mentioned above,the method performance,such as theresolution,trailing etc have all reached required level. The calibration curve was linear in the range of 0.16~0.8μg/mL (r=0.999993),The average recovery was 86.3%,In July leave had the highest content,which was 0.0115mg/g.In September seed had the highest content,which was 0.0307 mg/g.The method was rapid and accurate enough to determine the rotenone of Amorpha frutiicosa L,Simultaneously researched of toxicities effect of extract of Amorpha frutiicosa L on Hyphantria Cunea.the seed and leave extracts had higher contact toxity against Hyphantria Cunea,with contact LC50 values of 12.33 and 10.76μg/mL.The compatibility of extracts,Veratrum nigrum L and Veratrum nigrum L showed synergistic effect. Detect change of catalase (CAT) and peroxidase ( POD) activities in Hyphantria Cunea;respectively.35min after rotenone reatment,Compared to CK,the activity of CAT was no significance differences,55min after reatment,the activity of CAT increased.At the same time point,the activity of POD increased by 79% and 15% spectively,as compared with the control.150min after treatment,the activity of CAT and POD decreased;the seed extract of Amorpha frutiicosa L had great toxicity on Larvae.
