| To find a rapid and accurate detection method of Cryptosporidium, this paper compared commonly used for Cryptosporidium oocysts separation simulation methods, the results show that the Sheather's sucrose concentration is the best floating method. And the best detection method of Cryptosporidium oocysts in the commonly used methods is nested polymerase chain reaction (Nested-PCR), it has high sensitivity and great practicality.We used the appropriate elected method to detect Cryptosporidium infections of Siberia/Manchurian Tigers(MT) in Harbin and birds in Shanghai bird markets. One hundred and eighty-seven MT samples and twenty-nine other predacity felids animal samples collected were examined by Sheather's sugar flotation technique and modified fast-acid staining method ,the average infection rate was 1.07%(2/187),4.81%(9/187) and 3.4%(1/29),6.9%(2/29). But no positive purpose strip was found by Nested-PCR amplification. To detect the Cryptosporidium in two hundred and eleven birds fecal samples , twelve positive samples were detected by Sheather's sugar flotation combined modified fast-acid staining method, the positive rate was 5.43 %, after Nested-PCR amplification and test sequencing results shew that Cryptosporidium was Cryptosporidium baileyi.In this paper, some common Cryptosporidium oocysts collection methods and Cryptosporidium detection methods were reviewed by model experiments, then commented on them theoretically .The conclusions gave references to cryptosporidiosis rapid checks ,and how to choose a appropriate method in different laboratory conditions, this data is more reproducible and realistic. Through epidemiological and molecular biological investigations about MT and birds were infected by Cryptosporidium, the result provade a basis on prevalence and characteristics of Cryptosporidium, and made a great significance to prevent and control Cryptosporidiosis. |
