In this research,SRAP(sequence-related amplified polymorphism) molecular marker technique was applied to analyze the mapping population of Yellow catfish(Pelteobagrus fulvidraco) which contains 100 F1 individuals and their parents.A genetic linkage with High-quality was constructed by using mapping software MAPMAKER/EXP(3.0).The SRAP-PCR system was successfully established and optimized for Yellow catfish. And all of the 196 primer pairs amplified large quantity of genomic DNA loci which included 102 separation markers.Chi-square(χ2) analysis indicated that 85 separation markers conformed to the Mendelian ratio,and 17 makers showed segregation distortion.The genetic linkage map of Yellow catfish contains 75 separation markers which were located onto 11 linkage groups,and spans 1276.2cM in total.The length of each linkage group ranged from 78.6 to 162.1cM,and the number of markers per group varied from 3 to 11.The longest distance between adjacent markers was 57.3cM and the shortest was 8.2cM.Through calculation we got the value of expected genome length as 1605.4cM and the coverage ratio as 79.5%.Most of the markers evenly distributed in the linkage groups with less clustering loci.This result would be significant for the construction of High-density genetic linkage map with other kinds of molecular marker,QTL analysis of growth related characters and sexassociated marker isolation in Yellow catfish.
