It was significant to improve genetic progress that the excellent germ plasm of dairy cattle including embryo,semen and cattle were utilized in global, with the application and generalization of artificial insemination technique and embryo transfer technique.But concomitance the invasion of recessive hereditary disease to cattle was increased, especially that excellent bulls carried genetic defect gene spreading in global and causing significant monetary losses to the dairy production.A Holstein population with 515 individuals was genotyped by PCR-RFLP method to detect recessive hereditary disease (DUMPS,BLAD and CN). The cattle of different genotype were sequencing. All the results are as follows:1. UMPS gene 405 locus T/C PCR-RFLP results indicate: 405 locus T/C mutation causes an AVaI polymorphic locus and arginine/terminal codons transition. Three DUMPS-carriers WHICH THEY WERE two bulls and one cow were observed. In the Holstein dairy cattle and Holstein bull population, the percentages of DUMPS carriers were estimated as 1.17% and 1.27% respectively. Genotypic frequency of normal gene homozygote and mutant gene heterozygote was 0.9883,0.0117, and allelic frequencies of normal allele and mutant allele were 0.9942,0.0058.2. CD18 gene 383 locus A/G PCR-RFLP results indicate: 383 locus A/G mutation causes a Eco47I polymorphic locus and aspartic acid/ glycine transition.Three BLAD-carriers which they were one bull and two cows were observed. In the Holstein dairy cattle and Holstein bull population, the percentages of BLAD carriers were estimated as 0.78% and 1.27%, respectively. Genotypic frequency of normal homozygote and mutant gene heterozygote were 0.9922, 0.0078, and allelic frequencies of normal and mutant gene were 0.9961, 0.0039.3. ASS gene 86 locus T/C PCR-RFLP results indicate: 86 locus T/C mutations cause an AvaII polymorphic loci and arginine/terminal codons transition.Three CN-carriers which they were one cow and two bulls were observed. In the Holstein dairy cattle and Holstein bull population, the percentages of CN carriers were estimated as 2.53%, 1.55% respectively. Genotypic frequency of normal gene homozygote and mutant gene heterozygote were 0.9845, 0.0155, and allelic frequencies of normal gene homozygote and mutant gene heterozygote were 0.9922, 0.0078.
