Egg yolk antibody which is a kind of immune globulin in egg yolk isγ-livitin.It is named immunoglobulin of yolk(IgY) that antibody extracted from egg yolk in immunological domain,for it is difference to IgG extracted from mammal in physical property,chemical property and immunological property.Egg yolk antibody is composed with a lot of IgG and sparing IgM and IgA.To birds,hen transport immunoglobulin to chicken by ovum and protect chicken.This experiment is two parts.The first step is to establish a quantative double antibody sandwich ELISA,it include extraction of goose IgG(saturated ammonium sulfate method and Sephodex-G200),preparation of mouse anti goose antibody,preparation of rabbit anti goose enzyme labeled antibody,preparation of collatering negative serum(saturated ammonium sulfate method) and preparation of collatering negative egg yolk(dilution method and saturated ammonium sulfate method).Definition of test condition such as coating time, reaction time of sample and enzyme labeled antibody for ELISA.Sensitivity,specificity and reproducibility test for ELISA.After probate the ELISA,take the collatering serum and egg yolk as dilution,and add goose IgG in diverse density,then build goose IgG standard curve. The second step is to collect samples of Jilin white goose.The method of collect egg yolk is as fellows:collect 6 samples of egg yolk before hatching,collect samples every other day from 4-day-age,sampling 6 portion each time,the samples are diluted 7 times with sterile distilled water then keeped in -20℃refrigerator.The work of collect egg yolk is completed to the 34-day-age when the egg yolk disappeared.The method of collect serum is as fellows: collect samples every other day from 4-day-age,sampling 6 portion each time,the samples are keeped in -20℃refrigerator.After collected samples,detecting the serum samples and egg yolk samples separately with double antibody sandwich ELISA which established,then convert the detecting results to protein level with standard curve.According to the study,the tendency of IgG contents change is steady descend in the whole at the integral incubation period,and the volume of egg yolk is diminished correspondingly.This means that the change of total contents of egg yolk antibody is correlated with concentration of egg yolk antibody and volume of egg yok at the same time. Antibody content in egg yolk changed from 4311.196μg/ml before hatched to 2821.363μg/ml when egg yolk disappeared.The change of egg yolk volume is not very obvious before the 20 day-age.The volume of egg yolk decrease obviously after 22 day-age,so it is presumed that a lot of antibody is utilized by goose embryo at this period.The tendency of IgG contents change is upgrade in the whole,but the contents is comparatively low before 24day-aged,it hoick after 26 day-aged,and it reach to the peak,then decrease after 36 day-aged.
