| The experimental materials of this study were callus which were licorice(Glycyrrhiza uralensisf rsch),whose roots and rhizomes are the most common Chinese herbalm edicine,a kind of perennial suffurticosa herbal plants from Glycyrrhiza species Legume family,one of the traditional Chinese Medicines used to reinforce the function of the spleen and replenish.Besides medical value, the licorice is widely used in food,beverage,cigarette,cosmetics and other industries as additive.As natural resources can't fulfill require of medicine and other industries.Research of liquoice has been very hot in the world.Cultivation and tissue culture are considered to be hopeful way to alleviate the crisis of natural resources.Tissue culture technic is a good method,it can control grow condition of plants,accelerate pace of propagate,and distill effectual element from cells.Glycyrrhiza as material in the test.But for licorice tissue culture technology was not mature only few papers were reported,We study the course of callus effect of hormone to callus's uspension culture,observe the course of Defined the callus' state to translate Somatic embryo,offer theory and technic foundwork of propagation and distill effectual element from cells.The disseration reports genetic transformation in Glycyrrihiza by Agrobacterium rhizogenes and establishment of fast-growth Glycyrrihiza hairy root system as follows:1.Agrobacterium rhizogenes was applied to induce the Glycyrrihiza germ-free seedling,cotyledon,leaf,petiole with usual method.Comparing transformation ratio of different bacteria strains,we found that strain A4 is the best stain,and different sections of explants were infected differently.2.We had selected the culture medium which is best suitable for the growth of hairy root of Glycyrrihiza.Results show that MS is the very culture medium and 23℃is the best for the growth of hairy root.3.In suspension culture,cells grow curve show "S",The time of logarithim growth appears after inoculation 10-20 days;We have confirmed 25-30days is the time of gain hairy root cells.4.Polyphenoloxidase activity and Peroxydase activity is the highest in the green callus of the hairy root.5.IAA contents is the highest in the green callus of the hairy root.ABA contents is the highest in the yellow callus of the hairy root... |
