The sea cucumber species, Apostichopus japonicus, cultured in North of China Sea, is a valuable commercial species due to its social and economic properties. In recent years, the technique of sea cucumber aquaculture has been developed rapidly. However, the expansion and intensification of A.japonicus farming has led to the occurrence of some infectious and lethal diseases in 2002 , which makes people be aware of the fact that the aquaculture has come into a bottleneck . The complicated symptom of "skin ulcerative syndrome" has made people distract more and more interests to the biological characteristics of this cucumber. Thus, various means, such as morphological, physiological, histological and biochemical tests, have been applied in this field.Under the environment of artifical breeding, the early development of Apostichopus japonicus was observed and the morphological characteristic of each developmental stage was described. Embryogenic time-scale of Apostichopus japonicus was built and larval growth curve was protracted.The Sea Cucumber various systems and the construction of the organizational structure, which were observed and descriped comprehensively and systematicly, by using Anatomy, paraffin sections and H & E staining, and providing the theoretical basis and reference like for the further physiological research.Coelomcytes of Apostichopus japonicus were collected and purified as antigens to immune rabbit, and the preliminary optimization of producing polyclonal antibody to coelomcytes was explored, which gave a preparation to find coelomcytes homologous organs or tissues through further immunological method.Based on the indirect enzyme-linked immune response, the antibody titers were measured. It had a low value of 1:800, maybe due to the interference of viscera inside or bacterium in the environment. Using competitive ELISA test response, both Vibrio splendidus and Aeromonas hydrophila had little competitive ability to the coelomcytes, while the Organization of the respiratory tree had a certain competitive ability. It indicates there s a possibility of contamination by the respiratory tree in preparation of antigens.Indirect fluorescent immune method is used to detect the cells antibody. The cavity cells in the fluorescent microscope were found to combine the FITC tagged antibody as blue green cells, and with negative serum or without fluorescent antibody test observation show that fluorescent Weak or not to issue fluorescence. With fluorescent markers, respiratory tree shine with a light blue fluorescence which indicated that the antibody and respiratory tree have a certain combination.All the experiments above show that it is possible to establish an immunological method to find organs or tissues which have certain identity to the body cavity cells based on further biological technique to exclude bacteria and stichopus offal to produce high purity of polyclonal antibody to Coelomcytes, or use of monoclonal antibody technology to study histogenetical relations between these cells..
