Lycopene content was determined among 67 watermelons by spectrophotometry.A comparative study on lycopene content of different method,for example,sampling parts,extraction solvent,extraction temperature,extraction time,maturity and different storage condition.Study on lycopene content of different flesh color,different Ploidy,F1,F2 and Parents,the relations between sugar content and lycopene content were studied in this experiment.The results were as follows:1.A comparative study on lycopene content of different method,for example,sampling parts, extraction solvent,extraction temperature,extraction time,maturity and different preservation condition.A better method was found in this experiment.Firstly,sampling around seeds.Secondly,adding into 30mL n-hexane as extraction Solvent.Thirdly,the sample was oscillated about 2 hours without light in oscillator of 35℃,the rotation speed was 110r/min.Finally,absorbance was determined in wavelength of 503nm by spectrophotometry.2.A comparative study on lycopene content of different maturity watermelon,the results showed that: the lycopene content was slowly accumulated with fruit into a proper maturity stage,when watermelon was fully mature,the lycopene content was the highest,when watermelon was over-ripe,and lycopene content were also decrease gradually.The lycopene content of VS8-37 was the highest in the 36th day after fruit set and its content was 5.398mg/100gFW,the lycopene content of VS8-38 was the highest in the 34th day after fruit set and its content was 5.371mg/100gFW.Incised watermelon fruit were placed in dark condition Of 5℃and -18℃,storage time was 8d.The lycopene content reduced when the extension of storage time during the storage.The average lycopene conten decreased respectively by 23%and 31%in5℃and -18℃.3.A comparative study on lycopene content of different flesh color,different Ploidy watermelon,the results showed that:the lycopene content increased with flesh color deepened.The flesh color of ripe watermelon can be used as a apparent basis to determine the lycopene content of this period.Lycopene content of S292,S171-1 and S316 were higher than others,lycopene content were 6.372mg/100gFW, 6.135mg/100gFW and 5.978mg/100gFW respectively.The average lycopene content of 8 triploid watermelon were 6.315mg/100gFW,the lycopene content of 8 triploid watermelon were higher than their parents respectively,and their difference reached significance level.According to the calculated heterosis rate of triploid watermelon,the results showed that:the riploid watermelon had a positive mid-parent heterosis and positive transgressive heteriosis.4.The heterosis rate of lycopene content were calculated in three combination,the results showed that: there were two materials which showed positive mid-parent heterosis and positive transgressive in F1 of deep red flesh×deep red flesh;there were three materials which showed positive mid-parent heterosis heteriosis in F1 of deep red flesh×pink red flesh;there was no heterosis in F1 of pink red flesh×pink red flesh.5.Comparative study on the lycopene content of F2 of S649×S631,S649×S555P,S555P×S631, S649×S661,the results showed that the lycopene content ranged from 1.622mg/100gFW to 7.136mg/100gFW,which concentrated in 4.000~6.000mg/100gFW mainly.Broad heritability was 83.05%, 76.10%,79.89%,76.66%respectively.6.Correlation was analyzed in the sugar content and lycopene content of watermelon,The results showed that there was no significant linear correlation.
