TaAQP was cloned from Tamarix albiflonum and was transferred into plasmid pBI121 by sense and antisense. The sequencing showed that the received sequence proceeds were consistent with the target gene sequence. Agrobacterium tumefacience was transferred into tobacco by mediate transformation. Using PCR and PCR-Southern methods to detect the tobacco regeneration, the result showed that tobacco can receive TaAQP by sense and antisense successfully. By Northern detection, the gene can express, but foreign genes from different strains expressed different levels. All the foreign genes from four positive transgenic tobacco plants can express, and the number T1 express less, and the number T2, T5, T6 express more. In the four reverse-selected transgenic tobacco plants, all the foreign gene from number T1, T3, T5 and T8 can express. And the number T3 and T8 express less, and T1 and T5 express more.The protoplast was treated by hypotonic solution confirmed that the lines overexpressing sense TaAQP transport water faster than CK and the lines overexpressing antisense TaAQP. The protoplast from positive genes would numerously break within 5 minutes. But the protoplast from reverse-selected transgenic tobacco were treated by hypotonic solution, after 30 minutes, there are a certain number of intact protoplasts. As a result, the TaAQP gene function was verified.
