Chlorpyrifos is by far the most widely applied organophosphate insecticide,and its toxicity to human and organism has been paid more and more attention.To explore the effects of chlorpyrifs(CPF) on brain level of dopamine and organ coefficient in the offsprings of pregnant mice.The 7.5day and 13day pregnant ICR mice were randomly divided into 3 groups.CPF dissolved in dimethyl sulfoxide was administered by garage at dosages of 0,1 mg/(kg·d),5 mg/(kg·d) for 5 days.The amniotic fluid and telencephalon of GD13 foetus was separated,and the activity of acety cholinesterase in amniotic fluid and telencephalon was assayed; The brain of GD17 foetus,PN14 and PN60 mice s offsprings was separated,and then the brain was dissected into cortex,cerebellum, hippocampus,striatum which contained dopamine,dopamine level was analyzed.The results of 7.5day exposure indicate that CPF induced a fall in the activity of acety cholinesterase,and the inhibition rate of AchE was 80.4%and 73.7%in amniotic fluid and telencephalon ,but no toxicity symptom.The PN14 mice's organ coefficients of brain, liver and spleen were obviously different in 1mg/(kg·d);and The PN60 mice's organ coefficient of spleen is higher than control group,1mg/kg CPF induced a fall in the cortex of foetus and PN14 mice,but a rise in the cortex of PN60 mice;there was a rise in the cerebellum of three period;the hippocampus of PN14 and PN60 mice had also a rise;there was a obvious fall in the PN14 mice s striatum,but no variability in PN60 mice.The PN14 mice's organ coefficient of liver is higher than control group in 5mg/(kg·d);and the same results to PN60 mice's organ coefficient of spleen as PN14 mice.5mg/kg CPF induced a obvious fall in the cortex of foetus and PN14 mice,but no variability in PN60 mice; there was also a obvious fall in the cerebellum of foetus and PN60 mice;the hippocampus of PN14 and PN60 mice had a rise;there was a obvious fall in the PN14 mice s striatum,but a rise in PN60 mice.The results of 13day exposure showed CPF had no impact on PN60 mice's organ coefficient;The PN14 mice's organ coefficients of heart and spleen were less than control group in 1mg/(kg·d).1mg/kg CPF induced a rise in the cortex of foetus and PN60 mice,but no variability in PN14 mice;DA levels was less than control group in cerebellum of foetus,but higher in PN60 mice;the hippocampus of PN14 mice had a rise;there was a fall in the PN14 mice s striatum.The PN14 mices organ coefficients of kidney and lung were higher than control group in 5mg/(kg·d);but no variability in PN60 mice's organ coefficients.5mg/kg CPF induced a rise in the cortex of foetus,but a fall in PN14 and PN60 mice;there was a fall in the cerebellum of PN60 mice;the hippocampus of PN60 mice had a obvious fall;there was also a fall in the PN14 mice s striatum,but a rise in PN60 mice.
