PhospholipaseD(PLD) are catalytic phosphodiesterase key hydrolysis and bases of the floorboard of the exchange of enzymes.They are popular enzymes In the plant organizations.PLD and it's production PA play an important role in signal transmissions and plasmalemma degradations.In this study,we used two methods to search the PLD genes from the genome of the model plant,Arabidopsis and rice.Firstly,we performed multiple BLAST algorithms to search against the corresponding data set using the known PLD proteins got from several database as our query.Then,we analyzed the proteome data with a program written by Perl in which a regular expression was used to search for the PLD motifs from the genome data of the local Mysql database.We eliminated the proteins that did not contain the PLD motif after detecting by SMART and Pfam.Finally,we found 13 and 16 genes which encode the proteins containing the PLD motif from Arabidopsis and rice,respectively.A complete overview of this gene family was presented,including the classification of subfamily,chromosome locations,the analysis of the tissue expression,the prediction of the structure of PLD motifs.The main results are as follows:1. We used BLAST and the program written by Perl to identify 13 and 16 PLD family genes in Arabidopsis and rice,respectively.2. The phylogenetic trees of PLD gene family were generated with the neighbor-joining method.3. Based on the chromosomal location information on Internet,we localized Arabidopsis and rice PLD genes in chromosomes.We also appraisaled the sister genes and the homologous genes of the two species PLD genes. 4. We examined the expression of Arabidopsis and rice PLD genes using massively parallel signature sequencing(MPSS)and EST data. According to expression profiles,PLD genes can be classified into three groups.5. The microarray date from Genevestigator site showed that the expression of the Arabidopsis and rice PLD genes changed under stresses stimuli.6.We cloned a PLD gene from Arabidopsis named AT3G05630. the full-length of AT3G05630 cDNA consisted of 2,793 bps which contains an open reading frame of 2,434 nucleotides coding for a 811-amino acid protein.A sense vector named Prok2-PLD were constructed.
