The poisoning effect of ochratoxin A on liver in broiler is the object what we studied. 150 AA broilers purchased were fed from 1-day-old to 7 days . 100 broilers which have better growth conditions and have not significant difference in body weight were choosed. And theses broilers were randomly divided into four groups as the high dose, medium dose,low dose of ochratoxin A in toxic groups and the control group according to the addition dose of ochratoxin, 25 in each group. The broilers in toxic groups were injected with ochratoxin 12 ug.kg-1, 8 ug.kg-1, 4 ug.kg-1, respectively, and once a day in per broiler. The control group was injected with normal saline 1ml. The toxic groups and control group were injected continuously for 30 days. After the counteracting toxic substance, blood sampling in wing vein was used in the toxic groups and control group in 12h, 24h, 36h, 48h, 60h, and 72h time points, respectively. And the serum was separated, liver and kidney were anatomised from the chicken in order to studying some indexs as following:1. The affected of OTA on serum biochemical indicators of broiler chickens: the determination of active substances in serum, such as alanine aminotransferase ALT (U/L), aspartate aminotransferase AST (U/L), alkaline phosphatase ALP (U/L), total protein TP (g / L), glucose GLU (mmol/L), albumin ALB (g/L) and so on.2. the lipid peroxidation of OTA on the liver:determination of malondialdehyde (MDA), superoxide dismutase (SOD),glutathione peroxidase (GSH-Px), catalase (CAT ) enzyme, ATP enzyme, nitric oxide(NO)et al in liver tissue3. Histopathological changes of the liver.4. Liver cell apoptosis.The results showed that:1. With increasing of dose and prolonging of time,the activity of AST, ALT, ALP of serum increase , and ALB, GLU, TP of serum cut down.2. Whit increasing of dose of Ochratoxin A and prolonging of time, the activity of SOD,GSH-Px,CAT,ATPase in hepatic cells decreases gradually, but the content of MDA and NO increases gradually. Comparing with the control group, the difference of those six index was significant(P<0.05).3. OTA can lead to pathological changes of the liver tissue. Whit increasing of dose of Ochratoxin A,those toxins caused the hepatic sinusoid congestion, the permutation of hepatic cord inordinance , connective tissue hyperplasia; Increase in splitting the nucleus of liver cells increasingly split and hyalinize, lobules of liver have differential sizes, interlobular connective tissue thickening,cytoplasms are filled with a large number of build-up glycogen granules and many of the foreign bodys;In the sinusoid,some liver cells have been completely dissolved, endothelial cells have been proliferated remarkably,so the sinusoidal spaces and the gaps between the liver cells became smaller. Lesions increased with the increase in toxicity.4. Apoptosis rate of hepatic cells is increased with increasing of dose and prolonging of time.The result show that the increasing of weight of chicken is inhibited by Ochratoxin A. Ochratoxin A have toxicity for hepatic tissue and have influence for apoprosis program of normal cells. Death rate is accelerated by Ochratoxin A .
