In order to study molecular basis of heterosis of Liriodendron, the patterns of differential gene expression in plumules between hybrids F1 and their parents were analyzed by DDRT-PCR. The results were as follows.1. The total RNA obtained by the method of Trizol was good quality, high yields, and suitable for all downstream applications.2. There were obviously differential four gene expression patterns between hybrids and their parents. They were bands expressed in both parents but not in F1(Ⅰ), bands expressed only in F1 but not in both parents(Ⅱ), bands expressed in one parent and F1 but not in another parent(Ⅲ), bands expressed in one parent but not in F1and another parent(Ⅳ). In budding phase and bloom phase, proportions of 4 gene expression patterns in growth development stage of Liriodendron hybrids H×M were less than the proportions of Liriodendron hybrids BM×J. In dormant phase, proportions ofⅠandⅡin growth development stage of Liriodendron hybrids H×M were higher than the proportions of Liriodendron hybrids BM×J, but proportions ofⅢandⅣin growth development stage of Liriodendron hybrids H×M were less than the proportions of Liriodendron hybrids BM×J.3. There were significant differences onⅠandⅡamong differential growth development stage of Liriodendron hybrids H×M and BM×J. But There were no significant differences onⅢandⅣamong differential growth development stage of Liriodendron hybrids H×M and BM×J. There were significant differences on differential gene expression patterns (Ⅰ,Ⅱ,Ⅲ,Ⅳ) among budding phase, bloom phase and dormant phase of Liriodendron hybrids H×M and BM×J.4. By correlative analysis, we found thatⅢwas positively correlated with heterosis in leaf area,Ⅳwas negatively correlated with heterosis in basal diameter and plant height.
