Hedysarum mongolicum is one of the peculiar grazing in China. It is one of good materials which can be used in checking winds and fixing drifting sand, especially in improving the soil, changing the local conditions and improving the natural ecological environment. Utilizing genetic engineering to improve heredity of Hedysarum mongolicum has become an important practice meanings. Establishing an effective, steady system of tissue culture and transform system have become more important in molecular biological study and a key technology of genetic engineering of Hedysarum mongolicum in recent years.In this paper, we design and make the Multi-needle to Plate Equipment used for Negative Corona Discharge, and do research into the relation between different influencing factors and the characteristics of corona discharge. Meanwhile, the discharge current increase with increase of space between needle to needle, and the speed of the discharge-power' augmentation become tardy as well as the space between needle to needle is more than 20mm. Considering the stability and deferent power, 20mm space between needle to needle is suggested. In addition, the discharge current increase with reduction of space between needle to plane, and the deferent power of corona discharge is up to its maximum when the space is about 25mm.The experiment about the effects on callus treating with corona discharge show that corona discharge has certain influence on survival rate of callus. Based on the result of stating the water lose rates, weight increase rates and the test of calluscyte's survival rates, the dose of 14kv, 120min and 15kv, 90min for corona discharge is suggested.In the experiment of transformation, we get the resistant transformer from the test of GUS gene and filtration of kanamycin successfully and PCR analysis of the transformer of callus show that the soybean gene have been transferred into the Hedysarum mongolicum genome.
