A novel procedure was described for the hapten design of the N-methylcarbamate insecticide propoxur. Three new haptens of propoxur (Hapten-1a, Hapten-1b and Hapten-1c) were synthesized by introducing appropriate spacers in the pesticide aromatic moiety of the analyte molecular structure. First, the propoxur reacted with nitric acid to give the intermediate product. Then hapten-1a was prepared via the reduction of the intermediate product, hapten-1b and hapten-1c were formed by the acylation of hapten-1a with succinic anhydride and maleic anhydride respectively. In addition, we have also developed a simple method to prepare the hapten-2 for the pyrethroid insecticide cyhalothrin, which requires only two steps including the reactions of nitration and reduction. The molecular structures of these four haptens were identified by elemental analysis, IR, MS and 1H NMR.Four haptens were coupled to bovine serum albumin (BSA) and ovalbumin (OVA) to prepare the corresponding artificial antigens. Six new artificial antigens for propoxur and two artificial antigens for cyhalothrin were prepared, and those of the artificial antigens were confirmed by UV-vis.
