The experiment materials in this paper is two types of xeropHytic shrubs from Linze andMinqin,which name is Calligonum potaninii and Nitraria tangutorum Bobr.Using polyacrylamide gel electropHoresis technology, the isozyme analysis of the thirty-two individual functional leaves from Calligonum potaninii and Nitraria tangutorum Bobr is done,and the intrapopulation and interpopulation differences of genetic diversity and genetic relation of the two populations is analyzed.The conclusion obtained in this paper is shown below:1.The best deposition conditions and dyeing recipe for the electropHoresis test of Calligonum potaninii is put forward by the esterase isozyme analysis of this material.The key deposition condition is extracting buffer solution and 10% separation gel by using This-Hel.2. By the analysis of esterase zymogram of Calligonum potaninii,the significant difference shown on the esterase zymogram between the sixteen individual plants in each population is revealed,which can embody the polymorpHism of gene.The genetic relationship between the sixteen individual plants is indicated on the intrapopulation dendrogram,its genetic relationship is not very close and genetic differentiation is comparatively great.3. The intergroup dendrogram indicate that most of individual plants have the combined characteristics of syntaxis and divergence.therefore, a preliminary judgement is made that natural distribution and Self-perpetuation as well as natural growth is the main existing way of LinZe HeXi Calligonum potaninii.MinQing HeXi Calligonum potaninii is a group of artificial introduction. Its provenance came from the three areas above.LinZe group is one of the provenance of MinQing group is concluded by the zymogram information and dendrogram.4. The best deposition conditions and dyeing recipe for the electropHoresis test of Nitraria tangutorum Bobr is put forward by the esterase isozyme analysis of superoxide dismutase of this material.The key deposition condition is extracting buffer solution and 15% separation gel by using This-Hel.5. A certain extent of difference shown on the esterase zymogram between the sixteen individual plants of each population is indicated by a series of date and diagram from the zymogram analysis of superoxide dismutase of Nitraria tangutorum Bobr,but the difference is not sharp.The intrapopulation dendrogram indicate that the genetic relationship between the sixteen individual plants in each population is very close,which can embody the polymorpHism of gene.But there are some differences of genetics problem such as the number of Spectral bands and mobility ratio and so on.The interpopulation dendrogram indicate that the genetic identity between the two populations is high and the genetic relationship between them is close.therefore,theMinqin population should come from Linze population.
