Polyploidy is natural phenomena in plant evolution.In angiosperms,about 70%of the flowering plants have taken place once or more times of polyploidization in the evolutionary history.The direct effect of polyploidy brings out the genome duplicated, and changes in the genome of the double bound to affect the RNA expression of genes on the volume.Thereby,it affects protein expression,and ultimately affects the whole plant function and the traits expression.In this study,we select twin-seedling rice strain SARâ…¡-628,in which homologous naturally occurrs haploid,diploid and triploid.According to the analysis results of gene chip by Peng Hai,et al.,they did further analysis of the RNA of the expression level of the changes of single and triple-fold by real-time fluorescence quantitative PCR. According to the preliminary DNA microarray analysis,we chose five rice expression gene families of different functions for study,analyzed whether their changed locus in the expression is associated with ploidy regulation.Preliminary results are as follows:1 According to the preliminary findings of the selected chip 5 gene family function as follows:the formation of methylation associated with the site,MBD protein family, ethylene synthesis-related sites,SNF2 family associated with the gene regulatory sites and DNA repair related to the reorganization of spaces points,using fluorescence quantitative PCR to verify expression of 17 sites of gene family and found that:Results concordant ID of 76.92%was observed between quantitative PCR result and Chip result, but some sites of expression had variation compared with the result of chip.2 In the 9 sets of double rice seedlings 1N-2N-3N,there have 17 loci changed in expression level of these five gene families in the haploid and triploid(among of them,5 specific loci changed in the haploid,5 triploid specific loci changed,7 loci has changed expression simultaneously between haploid and triploid,results were as follows:1) At different times of rice in 108 sites,compared with the diploid,the sites of haploid expression increased,declined and had no significant differences were 22,63 and 23 respectively,the proportion were 20.37%,58.33%and 21.30%.And that of triploid were 97,2 and 9,the proportion were 89.82%,1.85%and 8.33% respectively.The results illustrate the expression tendency of these 17 loci in these nine pairs of seedlings was as follow:tend to down-regulated change after haploidization,tend to up-regulated expression after triploidization.2) The plants of same ploidy level had consistency at the same loci of the expression, but three loci in the expression showed no regularity among 9 haploids.3) A single locus expression changes could not affect the function of similar in the whole plant...
