Studies On The Extraction And The Non-specific Immunity Effect Of Ficus Polysaccharides In Crucian Carp(Carassius Auratus)

Plant Polysaccharides also being called Plant saccharans is extensively distributed in plants and bonded by some identical or dissimilar monosaccharide withα- orβ- glycosidic links,which has important biological activity.It has the effects of harmimegathy immunifaction,antisepticize,resist tumor,antivirus,reduce blood sugar,reduce blood-fat, antioxygen and other effect.The research inspected the immunoregulation effect of Radix isatidis polysaccharides,Schisandrae fructus polysaccharides,Ficus carica polysaccharides, and was concentrated on the extraction technology and the optimum dose of immunomodulatory properties of FCPS to be studied.The main results were as follows:Crucian Carp(Carassius auratus) were fed with the medicated feed containing polysaccharide from Radix Isatidis,Ficus carica,Schisandra for 21 days and analyzed for total serum protein level,phagocytic activity and bactericidal activity on day 0,7,14 and 21 by Coomassie brilliant blue method and MTT dying method to investigate the effect of these three polysaccharides on immunological function of Crucian Carp.The results showed that all of three polysaccharides can enhance the immune of the fish.There were significant differences in phagocytic activity and bactericidal activity(P＜0.01) compared with the control group;In the Ficus carica polysacchride and Radix Isatidis polysaccharide groups, however,there were significant differences in total serum protein level and lysozyme activity (P＜0.05);meanwhile,these three polysacchrides could reduce the dead rate of Crucian Carp infected with Aeromonas hydrophila.The optimal condition of extract by the method of digeratur experiment:the temperatue of extraction is 80℃,the extraction time is 6h separate with 2 times,the powder-to-water ratio is 1:9.The optimal condition of abstraction by the method ethanol precipitation:the powder-to-alcohol is 1:1,the end level of ethanol is 60%,the sediment time is 18h.According to the above-mentioned conditions,the rate of cute FCPS was 19.34%,the content is 69.7%.After laminar analysis from DEAE-Sepharose fast flow and dialysis the rate of FCPS-1(Ficus carica polysaccharides-1) is 0.134%,the content is 79.7%;the rate of FCPS-1 is 5.133%,the content is 93.4%.To study effect of Ficus carica polysaccharide on the ability of non-specific immune function in Crucain Carp(initial body weight 40±2g),four experimental diets A₁,A₂,A₃ and A₄ were formulated to contain 0.1%,0.2%,0.4%and 0.8%Ficus carica polysaccharide respectively,and the control(A₀) contained no Ficus carica polysaccharide.The influence of Ficus CaricaPolysaccharide was determined by examining their content of serum total protein and complement C₃,lysozyme activity,blood serum superoxide dismutase activity,bactericidal activity and phagocytic activity of leukocytes in 0d,7d,14d,21d,28d and 35d respectively.The results showed that the content of serum total protein and complement C₃,lysozyme activity,blood serum superoxide dismutase activity,bactericidal activity and phagocytic activity of leukocytes in group A₂ were significant higher than those in the control group(P＜0.05).The content of serum complement C₃,blood serum superoxide dismutase activity and phagocytic activity of leukocytes in group A₃ were significant higher than the control group(P＜0.01),and higher than group A₁ and group A₄ which reached to the top level on the 14d.The content of serum total protein and lysozyme activity in group A₃ were significant higher than group A₀(P＜0.05) which reached to the top level on the 14d.There were no significant difference between group A₂ and group A₃ on the content of serum total protein and complement C₃,lysozyme activity,blood serum superoxide dismutase activity and phagocytic activity of leukocytes,but group A₃ was better than group A₂.There were no significant difference between group A₀,A₁ and A₄ on the content of serum total protein and complement C₃,lysozyme activity,blood serum superoxide dismutase activity and phagocytic activity of leukocytes at other sampling time.The best indose is 0.4%.
