Male germ cells in vitro is a research hotspot in the biology of reproduction fields. At present, study on male germ cells in vitro in mammalian has made great progress.The process of spermatogoium's proliferation and differentiation and the process of meiosis from spermatocyte to spermatoblast have already been finished. But the mature of spermatid was still hard to be achieved. Reports on spermatogenesis in vitro in crustacean are less at present.According to the mammalian, cell culture of crustacean began lately and still failed to establish a permanent cell line. The tissues of in vitro were mainly haemolymph, ovaries and heart,but reports on testis in vitro in crustacean are less.In this paper, male germ cell in testis of Eriocheir sinensis as a research objective, on the basis of the data about cell adherence rate,cell survival rate and RNA/DNA ratio, the osmolarity and pH for male germ cell in vitro was tested. The results showed that the optimum conditions for cell culture in vitro were an osmolarity of approximately 1000 mmol/L. The favorable pH range of the male germ cell was pH 7.2-7.4.On this base, we observed the situation of growth in the cell culture and tissue culture.the results were found that the survival of time in the tissue culture was longer than the cell culture and the ability of proliferation was stronger.what's more, it could shape part monoplayer.In addition, base medium supplement with haemolymph of eyestalk ablation could enhance the growth of the male germ and could not promote the proliferation. The effect of 0 h,24 h and 48 h haemolymph had no signifisicince difference in the satistic level.
