Lilium regale,belonging to the family of Liliaceae, is distributed 760-2200 meters above sea level near Minjiang River in Sichuan province for the Lilium.It has many good characteristics,such as cold-resistant,drought-resistant,salt-tolerant,alkali-resistant,disease-resistant, high value for medicine;and has rich variations in colors and good fancy. It is so actual merit and significance for us to study Lilium regale, which is an excellent parent for hybridization.In order to identify and isolate correlative genes of flower colors and pollen, a petals cDNA library of Lilium regale was constructed. At the same time, its transcriptome was carried out. Then they were subjected to EST sequencing and bioinformatical analysis respectively. The main results were as follow:1) A high-quality full-length petals cDNA library of Lilium regale was constructed with SMATR cDNA Library Construction Kit.The quaity detection indicated that the primary library had a titer of 1.4×106, in which 95.83%clones were recombinant, insert sizes ranging from 1.0-3.Okb were obtained.and the length of inserts were more than 1kb. The results showed that this library obtained from our experiment was integrated and efficacious,which is suitable for ESTs sequencing and gene screening.2) In order to evaluate the quality of cDNA library and analyse ESTs,192 positive clones were randomly selected by PCR and sequced. Finally,184 EST sequences were obtained, and the average length was 496bp. All the 184 ESTs were analyzed, then, a total of 157 ESTs were obtained,including 13 different contigs and 144 singletons which were composed by only one EST. The redundant ratio was about 10.29%. After BLAST identity analysis,66 genes of known function and 118 genes of unknown function were obtained. All the effective ESTs were transformed into the format inquired by GenBank and submitted. Their accession numbers were from GT229409 to GT229581.3) 73501 ESTs from leaves, scales, roots and flowers of Lilium regale were obtained by Solexa sequencing technology, including 154 different clusters and 71958 singletons. The 11802 genes with homology alignment have been annotated different functions according to Gene Ontology three classification levels(the biological process.cell component and molecular function). According to the accession in COG database, genes with function annotation or putativefunction annotation were assigned to 24 categories.These genes were related to RNA processing and modification, chromatin structure and dynamics.energy production and conversion,cell cycle control, cell division and chromosome partitioning,amino acid transport and metabolism, nucleotide transport and metabolism.carbohydrate transport and metabolism.coenzyme transport and metabolism,lipid transport and metabolism, translation, ribosomal structure and biogenesis, transcription,replication. recombination and repair,cell wall/membrane/envelope biogenesis.cell motility,posttranslational modification, protein turnover, chaperones.inorganicion transport and metabolism,secondary metabolites biosynthesis.transport and catabolism,general function prediction only.function unknown,signal transduction mechanisms,intracellular trafficking, secretion, and vesicular transport,vesicular transport,defense mechanisms,nuclear structure, cvtoskeleton.
