Drought and other adverse circumstances can induces plants to synthesize some useful substances to protect themselves to grow. ABA and dehydrin are two of the most important substances, and they have some relationship in their synthesis. In this research, we detect the ABA contents and dehydrin gene wzy2 to analyze the relationship between they and drought, as well as the relationship between them.In this paper, we used plastic basins to plant four species of wheat, and sample them when the wheat grew to the three-leaf period, the sample time is 70%, 60%, 50%, 40%, 30%, 20% of the maximum soil moisture content, and one day, two days, three days after rehydration, detect the ABA contents and wzy2 expression. We also cloned the segment of the two genes wzy2 andβ-actin, which were used as the standard samples in the real-time quantitative PCR. At last, a kind of software was used to analyze the relationship of ABA contents and wzy2 expression.The conclusions of this research are as follows:1. We have detected the ABA contents of four wheat species by the method of ELISA. The ABA contents increase as the drought tolerance, the ABA contents would drop quickly after rehydration. In the same drought tolerance, the species which is high drought tolerance has the higher ABA contents, and and vice versa. The ABA contents and the wzy2 expression have the same trend with the changes of soil moisture contents. Besides, ABA can induce the synthesis wzy2, and they have some relationship which need to do some farther research.2. A part of wzy2 andβ-actin were inserted in the vector which was transform to the PMG109 to amplificate, then they would be used as the standard curve of the real-time quantitative PCR. We establish the method of real-time quantitative PCR to determine the expression of wzy2, and get an conclusion that the expression of wzy2 increases with the drought stress, and has a sharp drop after rehydrin. Different species have different wzy2 expression, in the same drought stress condition, the species which has high drought tolerance has the higher wzy2 expression.3. Analyzed the drought stress, ABA content and wzy2 expression by software. We found that the drought stress is an influence factor of the ABA content and wzy2 expression, and the ABA content is also an influence factor of the wzy2 expression, and more research need to be done to analyze their relationship.
