Zinc-finger protein PRDM16(which could activated a robust brown fat phenotype and stimulated brown fat-selective genes) could control the switch between skeletal muscle and brown fat. As an domainant contor factor for the brown fat cell fate, PRDM16 had potential use in the treatment of obesity. PRDM16 was taken as the candidate gene of regulate cattle growth traits and meat traits in this experiment.Genetic variations of PRDM16 genes were detected by DNA pool sequencing and CRS-PCR-RFLP techniques in 1045 individuals from four cattle populations (Jiaxian Red cattle, Nanyang cattle, Qinchuan cattle and Chinese Holstein), and association analysis were carried out to evaluate the effects of genotypes of the candidate genes on growth traits of three Chinese cattle (Jiaxian Red cattle, Nanyang cattle, Qinchuan cattle). The objects were to discovery the hereditary characteristics and to explore molecular markers with significant effects on economic important traits for efficient selection and improvement of Chinese cattle, and for the protection and usage of breed resource of Chinese cattle, to provide genetic information for foundation of molecular marker database. The results were as follows:The varations of seventeen loci (P1P17) including PRDM16 gene sixteen exons and 3'UTR were studied. In these seventeen loci, 10 mutations were first detected (NC007314.3: g.577 G>T, 614 T>C, 206966 C>T, 211563 T>C, 211804 G>A, 212237 T>C, 212234 A>G, 213772 C>G, 215683 C>T, 237110 C>G), the mutation g.577 G>T located in exon 2, the mutations g.211563 T>C, g.211804 G>A, g.212237 T>C were located in exon 9, others were located in the non-coding region. The mutation g.211804 G>A resulted to Ala(A)>Thr(T) of PRDM16 peptide, and g.212237 T>C resulted to Leu (L) >Pro (P) of PRDM16 peptide, g.577 G>T and g.211563 T>C were synonymous mutations. Three genotypes(GG,TT,GT) were detected in g.577 G>T mutation. And the frequencies of TT and GT were: 0.000~0.052 and 0.350~0.938. Three genotypes (TT,TC,CC) were found in the g.211563 T>C mutation, the frequencies of th TC and CC were: 0.087-0.691 and 0.000-0.177. Three genotypes(GG,AA,GA) were detected in the g.211804 G>A mutations, and the frequencies of the AA and GA were: 0.000-0.437 and 0.091-0.582. Three genotypes (TT,TC,CC) were found in the g.212237 T>C mutation, the genotype frequencies of the TT and TC were: 0.053-0.209 and 0.527-0.888. Four loci in the JX, NY and QC breeds were all morderate genetic diversity (0.25T,g.211563 T>C loci were completely linked, the g.577 G>T and g.212237 T>C,g.211563 T>C and g.212237 T>C were linked.In g.577 G>T mutation, different genotypes had significant association with chest width and withers height in JX population; different genotypes had significant association with body weight,average daily gain,body length,heart girth in NY population. In g.211563 T>C mutation, there were significant association between the growth traits (body length,withers height,waist corner width) and the different genotypes in JX cattle; TT and TC genotypes showed better performance than CC genotype in NY breed. For the 211804 G>A mutation, different genotypes had significant association with withers height,heart girth,body weight,waist corner width and hucklebone width in QC population; the hucklebone width was significant association with the genotypes in JX population; In NY population, at the 24 months, genotype AA were significantly higher than GG genotype in every growth traits. The g.212237 T>C mutation, different genotypes had significant association with heart girth,body length,body weight,rump length,hucklebone width in QC population; Withers height,body length,body weight,rump length,hucklebone width were significantly association with the genotypes in JX population; At the 24 months, different genotypes were significantly different in every growth traits in NY population.
