In this thesis, preadipocyte culture from subcutaneous (SC) and longissimus muscle (intramuscular, IM) adipose tissues were employed to investigate the effects of t10,c12-CLA on the proliferation and differentiation of preadipocytes and key enzymes involved in lipid metabolism between SC adipose and IM tissues in pig. There are two parts in this study:Part1: In vitro tissue culture system of SC adipose and IM tissues, and preadipocyte culture system taken from 30-days pig were constructed firstly. The experiment were divided into 2 treatments groups, 0.1 mmol/L bovine serum albumin(BSA, as control) , 100μmol/L t10,c12-CLA were added to the cultures for 10 d, six replicates were performed for each treatment. At the end of a 10-d treatment period. MTT assay, GPDH activity assay and oil red O staining were used to characterize the effects of t10,c12-CLA on the proliferation and differentiation of pig SC and (IM) preadipocytes in primary cultures. The results showed that: (1) No effects on proliferation of IM preadipocytes and SC preadipocytes were observed by adding 100μmol/L t10,c12-CLA (P>0.05). (2) Differentiation of SC preadipocytes were inhibited and IM preadipocytes were promoted by adding 100μmol/L t10,c12-CLA (P<0.05).Part2: In this trial, in vitro tissue culture system and experiment design were same to part1. Enzyme activities assays and PCR were used to study the effects of t10,c12-CLA on the lipogenic and lipolytic enzymes in SC and IM tissue. The results showed that: (1) Activities of HSL in SC adipose tissue and activities of ME and LPL in IM tissue were increased by adding t10,c12-CLA(P<0.05). (2) The expression of FAS in SC adipose tissue and the expression of HSL in IM tissue were decreased by adding t10,c12-CLA(P<0.05);by contrast, the expression of HSL in SC adipose tissue and the expression of LPL in IM tissue were promoted by adding 100μmol/L t10,c12-CLA (P<0.05).Collectively, these data suggested the effects of t10,c12-CLA on lipid metabolism of IM tissue adipogenesis differed from that on SC tissue adipogenesis at preadipocyte proliferation and differentiation and key enzymes activities and expression involved in lipid metabolism point of view.
