The colonization of gfp tagged strain of biocontrol bacterial strain ANTI-8098A in tomato plant was observed using laser scanning confocal microscope. The colonization concentration of ANTI-8098A in different parts of tomato plant was tested to make sure the endophytic colonization property of ANTI-8098A in tomato for the first time. The results showed that ANTI-8098A and its tagged strain could colonize in tomato plant especially in roots, from which the concentration re-isolated reached 8.80×104, 2.20×105 cfu/g. The concentration decrease in stems and leaves, which were 8.75×104, 6.25×104 cfu/g in stems, 6.60×103, 3.07×104 cfu/g in leaves.The effect of ANTI-8098A on the contents of chlorophyll and soluble protein, the activity of SOD, POD, PPO,and the rhizosphere microorganisms of tomato was also detected. The results showed that ANTI-8098A affected the contents of chlorophyll in a short time, the contents of chla, chlb and carotenoid were significantly higher than that of control after 5 days of treatment, there was almost no content difference on the 30th day. The chla/b value was significantly lower than that of control after 10 days of treatment. There were no significant effects to the contents of soluble protein and the activity of SOD and POD of tomato plant. The PPO activity of tomato roots was significantly promoted. The contents of bacteria, fungi and actinomycetes decreased in tomato rhizosphere soil after treatment with ANTI-8098A.The colonization dynamics of gfp tagged ANTI-8098A in different soil were tested with the dilution plate method and PLFAs which analyzed its affection to the microorganisms and the microbial community structure. The results showed that ANTI-8098A:pCM20 could colonized in different type soil for more than 160 days. The PLFAs analysis showed that the sum of PLFAs were higher than that of control and the PLFAs of bacteria, fungi, actinobacteria were also higher.The biocontrol effects of ANTI-8098A and its gfp tagged strain ANTI-8098A:pCM20 on tomato bacterial-wilt-disease was determined. The results showed that, both 108, 106 cfu/mL of ANTI-8098A and ANTI-8098A:pCM20 were able to postpone the developing of this disease, with a higher control effect. Each of correction affection of 108 cfu/mL of ANTI-8098A and ANTI-8098A:pCM20 was 100 % and 85.7 % and that of 106 cfu/mL of ANTI-8098A and ANTI-8098A:pCM20 were both 57.1 %.
