Saposhnikovia divaricata is a member of perennial Umbelliferae medicinal herbs. The predatory excavation, wasteland reclaimed and other reasons resulted in wild resources damaged widely. The progressive decline of wild resources and wind medicinal market demand becomes mainly conflict between supply and demand. Because advantages of the methods used in plant tissue culture production of medicinal plants, did not limited by regional, seasonal and climate condition, and growth rate of tissue faster. Therefore, according to the different stages of tissue culture conditions of culture were optimized to enhance the biological production of medicinal material, including the differentiation of bud induction, rooting culture, rooting for transplanting, and pharmacological ingredients of transplanting and transgenic hairy roots, the results as follows:First, under different conditions on the impact of adventitious buds, for example MS medium supplemented with 0.2 mg / L NAA mixed with 2.5 mg / L 6-BA and 0.4 mg / L KT respectively, and KT0.2 +6- BA 1.0 differentiation rate of S. divaricata were most high. Suitable for period of callus subculture was 25 d, and the number of continue subculture was 2. As the light intensity increasing from 5001x to 20001x, callus produced shoots increased, shoot robust, 2000lx was adapt to increase best in S. divaricata.Second, the impact of the rooting of tissue culture in S. divaricata, the rooting number was almost the same in three medium including MS, B5, and White, but there were differences in rooting, MS medium was highest. Then based on the MS medium to adjust elements of culture, the rooting number adding IBA medium was slightly more than NAA, with the low rate of rooting. By the standards of both root number and root rates, 0.4 mg / L NAA should be the the best hormone concentrations. By increasing Medium pH, shoot rooting show increased, pH 6.0 acts to reach the most suitable pH. Rooting rate from sucrose as carbon source was higher than glucose medium, rooting number presented conversely. Too much ammonia nitrogen is not conducive to the growth of tissue culture and rooting wind, when the ratio between NH4 + and NO3 was 1:4 acheatived the highest adventitious rooting Tested in several salt intensity, rooting rate and root number in two salt intensity, containg 1/2MS and 3/2MS were tiptop .Third, in terms of rooting medium on survival during tissue culture study of the effects, MS medium supplemented with auxin IBA, neutral pH, 1/2MS, MS with fructose as single carbon source or fructose as components carbon sources, and the nitrogen source completely from NO3- or NH4 + adjustment under such cultivation, could receive the maximum transplantation survival rate. submerging before transplantation could improve the survival rate of transplanted seedlings, under 2000 lx light and 15℃low termperture for root survival rate of transplant tissue culture induced the highest; other through the medium and water in balance 1/2MS 15d the wind training transplanting survival rate higher than the survival rate through the second. Cultured seedlings, the decision to transplant the main factor is the transplant survival matrix and transplanting environmental conditions. Under The soil mixed by Vermiculite, farmland and sand ( l: 2:1 ), survival rate of plantlets was highest, followed by sand and land (l: 3). And the degree of transplanting depth and stability were all influenced the survival rate of plantlets transplanted to a depth of 2cm and the compaction of soil cultivation method is more conducive to survival of transplanted wind. In the April wind cloudy evening the most appropriate transplant tissue culture.
