In this study,the protocorn-like body(PLB) was induced from leaves of fourteen species Phalaenopsis by the same medium, and this medium was also suitable for propagation of the protocorn-like body. The results show that the suitable medium for protocorn-like body inducement from leaves of fourteen species Phalaenopsis was 1/2MS+3mg/LTDZ+20g/Sucrose+ 3g/Lphytagel. The induction rate was:V31:32%,8106:33%,729:46%,777:51%,604:21%,602-3:9%,791:28%,8169: 24%,735:31%,607:21%,771:4%,608:38%,728-3:18%,788:1%. It provides the technical basis for large scale factory production of Phalaenopsis. And the medium components is very easy, low cost, good repeatability, subculture is efficient, greatly reduced the costs, increase profits.In this study, ICE1 is cloned from Arabidopsis's DNA, and connected to the 35s promoter in order to construct the plant expression vector OVERICE1,and construct the plant expression vector pCAMBIA2300YFP,transform it into tobacco leaf by Agrobacterium mediated method, demonstrate the plant expression vector of pCAMBIA2300YFP have the ability to express,provide technical reserves for transform the plant expression vector of OVERICE1 into Arabidopsis's PLB in the next step, establish a system for genetic transformation of Arabidopsis preliminary, and lay the foundation for obitan new varieties of Arabidopsis which have the cold genes.
