Cytoplasmic male sterility (CMS) is ubiquitous in nature as a maternal plant genetic phenomenon and which is an important basis in Crop Heterosis Utilization at the same time. Wheat as the main food crop, studying its utilization of heterosis and male sterility is the matter that many breeders have been trying to overcome. Many studies have shown that there are some direct relationships between mitochondrion and CMS in wheat, Exploration of the relationship between mitochondrial DNA (mtDNA) and CMS is very important in studyinging the mechanism of CMS in wheat at present.To explore the mechanism of CMS and the difference of male sterile lines with Ison ucleur and allocytoplasm in wheat, and provide theoretical and technical support for further identifying the wheat male sterile lines with allocytoplasm at the molecular level. In this study, three male sterile lines which nucleus is wheat 90-110, cytoplasm is respectively from kotschyi,bicornis and the T.speltawere were used as experimental materials,50 random primers were used to conduct differential expression analysis of the mitochondriai DNA (mtDNA) from three male sterile lines of Ison ucleur and allocytoplasm by RAPD technique..the main results were as follows:(1)3 special primers were screened,which can amplify repeatable special bands from three allocytoplasm male sterile lines.(2) A special band with 1500bp in length was amplified using the primer S22 in ms ms(Ae.ventricosa)-90-110. A special band with 1200bp in length was amplified using the primer Opaal6 in ms(T.timopheevi)-90-110. A special band with 670bp in length was amplified using the primer OPA05 in ms(Ae.Variabilis)-90-110.(3) The three Special bands had high stability and repeatability, it revealed an apparent Polymorphism in the three male sterile lines of wheat, and this polymorphism is likely to be the main reason of the difference of restoration in three male sterile lines. The three primers can be used as marker genes of mtDNA related to male sterility.
