Lily (Lilium spp.) is perennial herb, bulb type,monocotyledon. Lily by rich color, fragrance is pleasant, elegant and beautiful flower has been loved by the public. Especially in recent years with the development of national economy, China's flower market has become prosperous ,Lily demand increases year by year.In this study, a few good new cut lily flowers organs as explants, studied the impact of the embryo factors and technology system, established OT Hybrids Lily varieties, "Manissa" and Oriental hybrid lily varieties of "Siberia" somatic embryogenesis regeneration. Study made the following main results:1. Optimum choice of explantIn different parts of explants on callus had more significantly, filament was the most suitable explant material. Filament explants,high pollution rate,low pollution rate,induction time was short,inducting embryogenic callus .2. Optimization of callus induction medium"Manissa",filament callus induction, proliferation,NAA was more significantly(Callus F=109.4917**,Callusing percentage F=19.5731**).6-BA was significantly(Callus F=5.8604*, Callusing percentage F=5.4571*).NAA1.0 mg·L-1induced proliferation of best,6-BA2.0 mg·L-1 induced the best ,6-BA 0.2 mg·L-1 proliferation of the best."Siberia",filament callus induction,6-BA(F=7.3726**) was more significantly.2,4-D (F=4.5101*),NAA(F=3.7378*) were significantly.Filament callus proliferation,2,4-D (F= 5.660*),6-BA(F=5.0625*), NAA(F= 4.4782 *) were significantly.3. Embryoid body (somatic embryos) induced"Manissa" embryogenic callus induction,2,4-D(F=79.4807**),KT(F=38.4990**),6-BA(F=38.2930**)were more significantly.The best medium for MS + 2,4-D 1.0 mg·L-1 + 6-BA 1.0 mg·L-1 + KT 0.2 mg·L-1 (The proportion of embryogenic callus = 0.7905aA). "Siberian" embryogenic callus induction, 2,4-D(F=55.9290**),KT(F=33.1020**),6-BA(F =15.1952**)were more significantly.The best medium for MS + 2,4-D 2.0 mg·L-1 + KT 0.2 mg·L-1 + 6-BA 1.0 mg·L-1.Varietal differences in somatic embryogenesis, Induction of hormone 2,4-D> KT> 6-BA,but compared to Lily "Manissa" callus more embryonic cell proliferation strong. The somatic embryos into embryo culture, embryogenic callus into embryogenic capacity was 332.8/g,it can be successfully Lily globular, heart-shaped embryo, shield-type embryo, cotyledonary various stages of embryo growth of embryoids.4. Somatic embryos and plantletsIBA on somatic embryos and plantlets was significantly.The best medium for the MS + 6-BA 0.2 mg·L-1 + IBA 1.0 mg·L-1.5. Lily bulb formation and rootingLow concentrations of NAA (0.1 mg·L-1) is conducive to the increase of lily bulbs and root elongation.Help lily bulb formation and rooting ,MS concentrations were 1/2MS> 1/4MS> MS. The most suitable concentration of agar was at 7.0g·L-1 based on comprehensive conside- ration of plantlets growth, quantity and quality of the root developed.6. Acclimation transplantingDomestication after transplanting, seedling survival rate of 91.67%.
