Diabetes mellitus (DM) is one of the most common endocrine diseases. The mortality is only inferior to cardiovascular disease and malignancy in the developed country, and becoming the third fatal pathogeny. Diabetic nephropathy (DN) is a common but serious complication of NIDDM, and it's a main cause of death of diabetic. Natural anti-diabetic pharmaceutical are attracting attention in the medicinal field by the less toxicity and no-drug-resistance. The natural drugs are exploited rapidly in the developed country. sweet potato fol. flavone (FSP), extracted from the t sweet potato fol.is found to have bioactivities such as anticancer, antioxidant and hypoglycemic activities. However, very little is known about the effect of sweet potato fol. flavone in treating insulin resistance of NIDDM and preventing or delaying the onset of diabetic complications.FSP was extracted from sweet potato fol. by ethanol extraction and acetic ether abstraction technique. And the ingredient of FSP was obtained by separation and purification. The animal model of NIDDM was estabilshed by injection of alloxan. The mouse of NIDDM model were divided into four groups, including model group, FSP groups,Glibennia group and normal control group. We examined FSP effect on renal function and antioxidative activity of kidney in NIDDM rats. FSP significantly ameliorated diabetic renal damage and that related to enhancing the activities of antioxidant enzymes. The effect of was analysised by the experiments of hypolipidemia and test and the determination of weight and blood fat of NIDDM mouse.In order to research the effect of FSP on reducing blood glucose, ameliorating insulin resistance and renal protection in NIDDM mouse, the immunohistochemical stain was used to investigate the cell apoptosis, antioxidant ability. The results were listed as follows.1.FSP was extracted by the following procedure:deposition by ethanol, cleaned by AB-Macroporous resin and vacuum drying, and so on. Three FSP fractions were got by column with eluent of water and staning with Ag. The extraction rate of three FSP fractions were 0.446%; 0.441%; 0.443%.2.After FSP (high-dose) treatment for 40 days, body weight and the content of visceral fat mass were evidently decreased (P<0.01) The levels of TG, TC, LDL-c in FSP group were significantly lower, while their level of HDL-c increased significantly (P<0.05,0.01), which implied that FSP has the function of hypolipidemia. The results indicated that FSP could protect the liver of the diabetic rats. Furthermore, HbAlc was lower.3.The toxicity and side effect of FSPwas examined by HE stain,acute toxic test Results showed that FSP could increase the quantity of the pancreatic islets and the cells in pancreatic islet; FSPhas the ability to amend the impairment on and kidney of allox-induced diabetic rats.4.Effect of FSP on apoptosis was studied; the results showed that expression of Bax By immunohistochemical analysis, the results implied that FSP also ameliorated expression of Bax and Bcl-2 protein in liver, and restore the function of saccharide and lipid metabolizing, consequently, which avoid diabetic complication.In the study, effect of FSP on insulin resistance in NIDDM mouse systematically and discussed the action mechanism from much point of view. It provided comprehensive, scientific evidence for development of sweet potato fol. flavone as a suitable dietary natural antidiabetic agent.
