Botanical pesticides WCT (polyhydroxy Dinaphthaldehyde) was selected from the Malvaceae plants,which was a viral replication inhibitor. Apple canker disease commonly known as rotten bark disease, smelly skin disease, is an important apple disease in northern China, which also result in weak trees, dead branches, dead trees, or your garden was destroyed completely. To explore the antifungal activity and physiological pathological effects of WCT against Valsa mali, the results can be listed as follows:Agar plate tests was conducted to test the inhibitory effect .WCT displayed strong antifungal activity to V. mali, the inhibitory was 77.4%. Jun duqing was 83.8%, Yu heling was 63.4%.While the concentration of WCT was higher (over 50mg/ml to liquid), the inhibitory effect was better. In the field, apple canker disease was treated by WCT, with an average healing rate of 40.7%, with an average efficiency increase of 66.1%.Apple trees was treated by WCT one year later, which scars can heal a wide and thick layers.The activities of PPO and POD and the contents of chlorophyll in apple increased significantly after induced treatment with SA and WCT, which was higher than check. In the 7th day, POD activity and chlorophyll with SA and WCT treatment reached to the highest point. Previous results showed that the PPO and POD activity was positively correlated with disease resistance of plants in general. But different enzymes and different plants reached to the highest point was at different times. In this study, the results were second and seventh day.The results of SDS-PAGE indicated that WCT and SA could induce a specific acid-soluble protein about 59KD in the 7th day express more than check. 59KD protein expressed higher on apple leaves with inoculated with V.mali, treatment than healthy plants. It indicated that this protein is produced by host plants and it was pathogenesis-related disease with apple canker. The obtained peptide mass fingerprinting (PMF) was searched in the protein data bank: swissprot, indicating that it was ribulose 1, 5 - bisphosphate carboxylase.Samples based on low molecular weight protein relative mobility and molecular weight Mr logarithmic Log Mw standard curve to calculate the protein molecular weight of this protein was about 59.Purified 59 KD proteins and agar plate tests within the solution experiments, we found 0.38mg/ml has greatest influence on V. mali. The result of Western blotting indicated that healthy plant and plant inoculated with V. mali expressed less protein than induced treatment with WCT and SA.
