| Salt stress seriously affects plant growth and development.Screening and identification of salt tolerant gene is the basis for salt tolerance breeding.Salt stress is controlled by multiple genes.Transcription factor has played an important role in plant growth,development and its response to the external environment stress.After screening the membrance containing 526 wheat transcription factors from our home-made wheat full length cDNA libraries with the probe under salt stress,two candidate transcription factors related to the salt tolerance were obtained.TabZIP 60 is 1741bp in length,encoding 338 amino acids. The homology analysis showed that TabZIP 60 had the very closer relationship with the A family of Arabidopsis and rice. After high salt treatment and ABA induced,the expression of TabZIP 60 was increased. Under PEG and cold stress-induced,its expression pattern was different from the salt stress. Subcellular localization of GFP showed that the transcription factor was a nuclear localization protein.TaC2H2 11 is 2496bp in length,encoding 613 amino acids. After salt treatment,the expression was increased. Under ABA,PEG and Cold stress induced,it showed the different expression patterns from salt stress. Subcellular localization of GFP revealed that the transcription factor was located in the nucleus.In order to validate their function,two expression vectors were constructed using Gateway technology,and transformed the two transcription factors into Arabidopsis. Functional analysis is underway. |
