Study On Biological Characteristics And Tissue Culture Technique Of Rare Ornamental Plant "Haierlian"

"Haierlian"belongs to Illiciaceae plants which was introduced to Suzhou from Yunnan, with a 300 years history of culture up to now. But its existing plants is so few that"Haierlian"is called Rare Plant Resource. At the same time there is little study about its ecological habit, status of species classification, method of vegetative propagation and so on. So in this thesis biological characteristics and genetic diversity of"Haierlian"with Illicium laceolatum A. C. Smith were studied by morphology search and molecular technology, while plant tissue culture and its application feasibility of rapid propagation in"Haierlian"were discussed. The results below were obtained mainly.1. It was investigated and analysed on the morphological characteristics, growth and development, Environmental effects and landscape application of rare plant,"Haierlian"in Suzhou. Their biological and ornamental characteristics were studied systematically. It was clear of its growth and development regularity of branch, leaf and flower."Haierlian"and Illicium laceolatum A. C. Smith could flower twice in one year. Some attentional issues were proposed about landscape greening and cultivation technique.2. In order to identify genetic diversity and genetic relationship of"Haierlian"and Illicium laceolatum A. C. Smith, molecular biology experimental methods were used to analysis genetic diversity between"Haierlian"and Illicium laceolatum A. C. Smith on DNA level and probe subclass classification between them. And differential expression was studied through proteome experimental technique. Genetic diversity was detected abundantly in leaf proteome between"Haierlian"and Illicium laceolatum A. C. Smith and it was determined preliminarily to be Illiciaceae, Illicium Linn., Illicium laceolatum A. C. Smith.3. As there is no seed and the natural breeding of"Haierlian"is very difficult, up to now there is no immediate reports of successful reproduction. Therefore, basing on the tissue culture the present research was carried out to study the vitro culture technology of"Haierlian"and establish the sterile tissue culture system. Technical conditions about tissue culture of"Haierlian"were selected from the choice of explants, the primary medium ,the subculture medium and so on. Finally appropriate condition were obtainted, and the test-tube seedings in vitro culture were got under above congdition. These lay the foundation of theory for the tissue culture of"Haierlian".
