| The male New Zealand White Rabbits were immunized 2C′3AB protein expressed from E.coli emulsified with Freund's adjuvant,201 adjuvant and 206 adjuvant respectively to produce polyclonal antibodies. The titers of the rabbit immune sera were measured by ELISA, moreover ,the purity, specificity and sensitivity of the polyclonal antibodies were identified by SDS-PAGE, ELISA and West blotting afer purification. The results showed that the adjuvant effects were 206 adjuvant>Freund's adjuvant>201 adjuvant. In addition, the polyclonal antibodies could specially reacted with FMDV positive serum with high purity and The titers of polyclonal antibodies were higher than 1:51200.To develope a rapid and accurate test for detecting FMDV nonstructural protein antibody based on the principle of gold immunochromatographic assay ( GICA).The FMDV non structural protein 2C′3AB inclusion body was conjugated with colloidal gold, which was called gold probes and was dispensed on the glass fiber. The 2C′3AB inclusion body and the rabbit anti 2C′3AB IgG were separately sprayed on the nitrocellulose membrane as the test line and the control line. The rapid gold immunochromatographic strip was assembled by the different accessory in regular sequence.If there was 2C′3AB protein in sample, gold probes on the glass fiber and 2C′3AB inclusion body formed a complex with serotype 2C′3AB protein,which was immobilized , furthermore , concent rated and appeared a distinct red color on the test line. In the clinic test , a total of 329 field samples were comparatively detected with both GICA and 3ABC indirect ELISA , and the coincidence rate was more than 85%.The evaluation test proved GICA established in this study is simple , rapid , specific and sensitive. It is suitable for differential diagnosis of FMDV.
