Since natural Calculus Bovis are of low yield of as well as much weaker pharmacological action of artificial Calculus Bovis compared with natural one, many domestic researchers have succeeded in cultivating Calculus Bovis in vitro and in vivo, but the output and the quantity of natural Calculus Bovis was not stable enough, may be because they only focused on the yield and neglected the formation mechanism. Through these trials of formation of Calculus Bovis in the external Calculus Bovis generators, the effects of E.coli,tropina,β-G and ConABP in forming gallstone during the process of formation were studied, and the changes of the key components of bile were dynamically observed.Trial 1 (The effect of E.coli,tropina andβ-glucuronidase in forming gallstone)Tropina andβ-glucuronidase were in the crushing liquid after ultrasonication of the E.coli which was acclimated, and theβ-glucuronidase was inactivated by 64℃water bath. Different densities of inactivated bacterium liquid (inactivation groups), crushing bacterium liquid (crushing groups) and E.coli (viable organism groups) were used to study the formation of bezoar in the external calculus bovis generators. Results indicated that: The visible surface areas of the Calculus Bovis nucleus of crushing groups was the largest among the three groups .The viable organism groups had an early forming time.Trial 2 (The effect of Con ABP in forming gallstone)ConABP was extracted from bile and gallbladder parietals of cattle by chromatographic affinity, and further analyzed by electrophoretic analysis. Different densities of ConABP were used to study the formation of bezoar in the external calculus bovis generators.Results indicated that: 0.0109mg ConABP can be extracted from 1 mL bile of cattle.ConABP increased both the quantity and visible surface area of the Calculus Bovis nucleus. The visible surface area of the Calculus Bovis was biggest when 0.0207mg ConABP in 1 mL bile, while the quantity of the Calculus Bovis nucleus was most when 0.0109mg ConABP in 1 mL bile. Trial 3 (The effections of Compound groups in forming gallstone)Different densities of inactivated bacterium liquid (m compound group) and crushing bacterium liquid (p compound group) were combined separately with 0.0207mg/mL ConABP, and used to study the formation of Calculus Bovis in the external calculus bovis generators. Results indicated that: 0.1×108cfu/ mL inactivated bacterium compound with ConABP had promotive effects on both quantity and visible surface area of the Calculus Bovis nucleus at the beginning and middle time of the formation. The maximal quantity of the Calculus Bovis nucleus from all the combination groups in p compound group was lower than the same concentration groups of m compound group.
