Strains are one of the important means of production in mushroom industry, and its quality directly reflects the development of mushroom industry. In this thesis, three Flammulina velutipes strains including F0025, F0030 and F0305 are studied from strains purity, strains fingerprinting and strains activity, which are used to establish complete strains quality control technology. Main results are as follows:1.Strains PurityThree strains of mycelium are used as material to tested from bacteria, mold spores, endophytic bacteria, viruses respectively. Bacteria are detected by means of streak plate, plate coating, and boiling as well as liquid culture for DNA PCR extraction. A limit can be reached with each method by manipulation. Fungal spores are detected by plate coating method to the limit of 100 spores per gram of strains. Endophytic bacteria is not detected by means of protoplasts blasting and antibiotic treatment. dsRNA is detected in the viruses by means of dsRNA technology and validation while not detected in other three Flammulina, which shows that this method can be used for edible virus detection, and three kinds of Flammulina used in the experiment are not contaminated by the virus.2. Strains FingerprintingFrom the species and varieties within the two levels of the mushroom for identification. By ITS-RFLP technique can distinguish edible mushroom species and open to the other. Re-use DNA molecular marker techniques in species identification of Flammulina. Taking into account the molecular marker SRAP compared with other simple, stable and advanced features of polymorphism.In this experiment, three kinds of mushroom SRAP amplification, the results screened 22 pairs of SRAP primers, obtained 33 polymorphic bands, identification accuracy rate of 99.99%.3. Strains ActivityThree strains of mycelium at different culture time are used as material to measure several physiological and biochemical indicators related to strain aging with results showing that Flammulina velutipes with different culture time were no significant internal links with CMC enzyme, xylanase, amylase and TTC enzymes; while polyphenol oxidase in different mycelia is gradually reduced with the increasing of the ageing time ,so it can be used as a measure dynamic strain indicator. In the high temperature resistance experiment, with increasing age of strains high temperature capacity of bacteria decrease or disappear. Therefore, high temperature capability can be used as an indicator of dynamic detection of bacteria.
