According to "Flora algarum sinicarum aquae dulcis", Enteromorpha is classified in Chlorophyta, Chlorophyceae, Ulvales, Ulvaceae. Enteromorpha is distributed broadly in the world, which is adaptable, from the high salinity field to fresh water rivers and from the hot equator to the cold Arctic. Enteromorpha has high food and medicinal values that has been found and used by masses of working people long ago. The genus Enteromorpha is used in the experiment commonly, and it is used as a model organism for the research of algae physiology. Enteromorpha was found very early, but because of its simple morphology and high variability, its classification status is disputed constantly and changed for several times from birth. With the development of molecular biology, it provides a new method for the classification of Enteromorpha to adopt molecular methods combined with morphology.In this paper, a freshwater Enteromorpha collected in Shanxi province as research material is observed under the microscope, extracted DNA and amplified rbcL gene,18S rDNA gene and ITS gene. Combined with Genebank sequences, the systematic phylogeny relationship between this Enteromorpha species and other green algae in Ulvales was researched by gene sequence analysis. The phylogenetic trees were established by neighbor joining, maximum parsimony, maximum likelihood and Bayesian methods together with the morphological study in the genus Enteromorpha. Advantages and disadvantages of the various methods and application range were discussed, and the results were compared. We considered that this species had a closer relationship between Enteromorpha linza,Enteromorpha intestinalis and Enteromorpha prolifera.The study did not determine the exact status of this Enteromorpha, so needed further research combined with more samples. The study also found that Ulva and Enteromorpha were not strictly differentiated. Therefore, more proofs were needed to give a better support of the right classification between the two genera.
