Brassica napus is one of the important oil crops in our country, rapeseed oil accounted for 40% of annual edible oil every year. The quality requirements of rapeseed oil increase with the improvement of people living standard. Rapeseed oil with high oleic acid content is considered to be beneficial to people's health, it still shows great advantage on medical, industry, and daily life, so, it's the important way now to increase the content of high oleic acid on rapeseed quality breeding.In this paper, we made further explore the function of acid-related genes as the starting point, took genetic engineering technology as a primary means, under the principle of RNA interference, picked up two gene pieces from the oleic acid rapeseed mutant subtracted library which we made in our laboratory (we found they had different expression pattern in high and low oleic acid material by RT-PCR analysis), we made the construction of RNA interference vector, and made the transformation of Brassica napus XY15 by Agrobacterium tumefaciens.1. Amplifying two desired fragments B34 and B35 from the leave DNA of Brassica napus XY15 by primers designing. we inserted the pFGC5941.Na carrier with NapinA promoter forward and reverse separated in both this two fragments, two RNA interference vector was constructed-pFGC5941.Na-34 and pFGC5941. Na-552. Interference vector was transformed into Agrobacterium. We selected cotyledons with petiole which was from Brassica napus plantlets as explants, which were transformed by Agrobacterium infection, screening for resistance in the induced buds after infection by PPT, obtained the optimum concentration was 1.8‰.
