This research investigates the omentin and visfatin mRNA in relative expression of different muscle and adipose tissues in Landrace pigs (a leaner, Western breed) and Rongchang pigs (a fatty, indigenous breed) and analyses the effect of gene expression in different breeds and tissues on fat deposition by using real-time quantitative PCR technique (SYBR(?) Green I method). The automatic biochemical analyzer and enzyme-linked immunosorbent analysis (ELISA) are used to detect the circulating levels of biochemical index and obesity-related cytokines, and the correlation between the circulating amount of these cytokines and the Volume value of the POI (Porcine obesity index) has been also analyzed.The results reveals that:(1) Volume value is more accurate than POI in the same breed and growth environment for the measurement on porcine obesity, more reliable conclusions should be gained by combining body mass index. (2) Detection result of the various circulating cytokine amount shows that TNF-α, IL-6 and LDL level in the more fat deposition individuals is higher than individuals with less fat deposition. The GHRP amount of Rongchang sires which are in the situation of growth phase and whose weight is extremely significantly lower than Landrace pigs and Rongchang sows is extremely significantly higher than Landrace sires (P<0.01). The LDH and LR amount of the Landrace sows are extremely significantly lower than Rongchang sows, and their VLDL amount is significantly lower than Rongchang sows (P<0.05), but the TNF-αamount is extremely significantly higher than Rongchang sows (P<0.01). The LDH and LR amount of the Rongchang sows are higher than Landrace sows but reverse in TNF-α. The amount of TC, APOB, LR, IL-6, GHRP-Ghrelin, Orexins and TNF-αin the serum of the Landrace sires are positively correlative with Volume value, which is negative correlative with SS. GHRP and leptin of Landrace sows are the negatively correlative with Volume value, and TNF-a, C-peptide and ADPR are positively correlative with Volume value; the correlation between the amount of LR, IL-6 and Volume value is significantly positive (P<0.05). TNF-α, Orexins and insulin of Rongchang sires are positively correlative with Volume value, which is negatively correlative with GasR and ADPR. The relationship between TNF-a, PAI-1, leptin and Volume value reveals that the adipose amount of the Rongchang sows is obviously accumulating, whereas the amount of C-peptide is abnormal. (3) Between sires and sows in the same breed, omentin mRNA of pericardial in sires is extremely higher than sows, and similar in abdominal fat and lesser omentum. But omentin in leaf fat and intramuscular fat of sows is extremely significantly higher than sires (P<0.01), and similar in greater omentum, lesser omentum and psoas major muscle. The omentin mRNA of upper layer of backfat and inner layer of backfat are reverse and extremely significantly different (P<0.01) between sires and sows of Landrace and Rongchang pigs. The result of comparison in different breeds of the same sex reveals that omentin of leaf fat in Landrace pigs is extremely significantly higher than Rongchang pigs (P<0.01), but in both muscle tissues, the result is reverse (P<0.01). The result of various tissues of different breeds shows that omentin of leaf fat and greater omentum of Landrace pigs are extremely significantly higher than Rongchang pigs (P<0.01), whereas Rongchang pigs'in intramusclar fat, inner layer of backfat, psoas major muscle and longissimus dorsi muscle are extremely significantly higher than Landrace pigs'. (4) The comparison result in different sexual pigs of the same breed reveals that visfatin in greater omentum, abdominal fat, leaf fat and intramuscular fat of sows in both breeds are extremely significantly higher than sires (P<0.01), except for greater omentum and inner layer of backfat of Rongchang pigs. The visfatin of lesser omentum in sires is extremely significantly higher than sows (P<0.01). In upper layer of backfat of Landrace pigs, the visfatin of sires is higher than sows, but sows' is significantly higher than sires'in Rongchang pigs (P<0.05). Between both breeds of the same sex, the visfatin of inner layer of backfat in Landrace sires is extremely significantly higher than Rongchang sires (P<0.01), and the expression of Landrace sires'visfatin in greater omentum, leaf fat and inner layer of backfat is higher, but Rongchang sires'is extremely significantly higher in longissimus dorsi muscle and lesser omentum than Landrace sires'(P<0.01). The visfatin of abdominal fat, greater omentum, leaf fat and inner layer of backfat of Landrace sows are extremely significantly higher than Rongchang sows (P<0.01), whereas the result is reverse in longissimus dorsi muscle, intramuscular fat, upper layer of backfat and lesser omentum (P<0.01). The comparison result of various tissues between both breeds shows that the visfatin of leaf fat, greater omentum, abdominal fat and inner layer of backfat in Landrace pigs are extremely significantly higher than Rongchang pigs (P<0.01) by 2,5,1.5,3 fold respectively, and visfatin expression of Rongchang pigs is extremely significantly higher than Landrace pigs in intramuscular fat, lesser omentum and longissimus dorsi muscle (P<0.01).
