Goose parvovirus (GPV) infection known as goose plaque, belongs to Parvovirinae Dependovirus, and causes high mortality in domestic goslings and Muscovy dueklings. It caused 90% mortality in susceptible goslings under the age of 4 days to 20 days. It can cause acute enteritis and also liver, kidney, heart and other organs septic lesions, particularly the fibrous and embolism of the small intestine. At present, GPV infection spreads every year in different degree that hinder the healthy development of breeding industry. Owing to the hinder the breeding industry development because of this desease, there needs accurate,convenient diagnostic method for early diagnosis.The technique of colloidal gold immunochromatographic assay just is a simple, easy and allegro immunology detection technology. It has been successfully applied to many domains, as to detect hormone or protein relate to disease, protein relate to pathogenic microorganism or antigen and antibody, the antigen and antibody relate to parasites, drug concentration from blood and urine since Beggs used it detect human chorionic gonadotrophin (hCG) at the early of 1990s. But there still is not the report for detection GPV antibody utilizing the technique now,however,it will bring about important influence to the healthy development of goose breeding industry that the technique is used to rapidly detect GPV antibody due to its advantages.GPV-VP3 is viral main capsid protein, approximately to seize eighty percent of capsid protein total count,and amino acid sequences that are exposed to the virion appearance are all show up inside VP3 polypeptide chain,what's more ,VP3 contains GPV main antigenic determinants,is the major immunology protective antigen,can have a specific binding with the antibody from natural infection goose,so VP3 could be labeled by colloidal gold and capture antibody.In this study ,a colloid gold strip (CGS) test for detecting antibodies to Goose Parvovirus (GPV) was developed. 18nm colloidal gold was prepared with the methods citrate sodium- chlorauric acid redox process .The test is based on membrane chromatography and uses colloidal gold conjugated with recombinant protein GPV VP3 as the tracer.On the test strip, the immune globulin rabbit anti-goose IgY polyclonal antibody was used as the capture complex at the"test line", and rabbit anti-GPV VP3 polyclonal antibody was used as the capture antibody at the"control line". and with the Point-of-care testing (POCT) scanner, GPV antibodies was quantitatively studied and the lowest level of antibody is 9.89 ng/ml. Compared with the agar gel diffusion test and the enzyme linked immunosorbent assay, the coincidence of the CGS test were 100% and 97.7%, respectively. Because the test strip is high specific and sensitive, convenient, rapid and good repetitive, the CGS test should be useful for detecting GPV antibodies in the field, monitoring the antibody level and its changing regularity of the goose, be easy to undertake immunologic protecting at the right moment, so it will grow down the damage to goose breeding and be in favour of the healthy and stable development of goose breeding.
