The encoded protein of BMP-6 gene is one of bone morphogenetic proteins. This kind of proteins belongs to the ransforming growth factorβfamily. Members of this family can regulate differentiation of Osteoblasts and Chondrogenic, Induce the formation of ectopic bone, promote bone healing and overmaster of the formation of different morphological characteristics mammalian bone in vivo or in vitro. With a detailed study, they found the biological function of BMPs family is not only limited to development and the formation of bone, but also involved almost all body system, including cardiovascular, gastrointestinal, respiratory, urogenital, hair.also involving growth and differentiation of nervous system, which make BMP-6 gene get more and more attention in human medicine and animal breeding field. Intention of this study is understand and explain the construction, expression and affect of reproductive traits of BMP-6 gene in sow.This study first cloned the full-length of BMP-6 CDS sequence of swine, analyze its structure function and molecular evolution by bioinformatics. Detect the polymorphisms and approach the distribution of different allele by PCR-SSCP technology, and study distribution of different genotypes in swine, and what affection they done about litter size and number born alive in sow. By using Real-Time PCR, study expression of BMP-6 gene in different tissues. The main results are as follow:1. For the first time, we cloned the full-length of BMP-6 CDS sequence (Genebank NM001168001). Full-length of swine BMP-6 gene is composed of 1518bp encoding 505AA.2. Two polymorphisms found in CDS sequence of swin BMP-6 gene by using PCR-SSCP. One of them, which detected by primer BMP-s206 and BMP-a207, is synonymous mutation, Another, which detected by primer BMP-s215 and BMP-R, is mutation wrongly italy.3. The polymorphism, which detected by primer BMP-s215 and BMP-R, is associated with litter size and the number born alive. there were significant difference among Wild boar and Yorkshire and French Yorkshire (P<0.01), difference between Wild boar and Landrace (0.010.05)4. By study correlation between polymorphism of BMP-6 gene and reproductive traits, we found positive affection. AB type get the highest numbers of piglet, average 13.00 pigs, average 6.71 pigs more than BB type, which is lowest, difference is significant (P<0.01).For number born alive, AA type get the highest, average 10.81 pigs, average more 3.69 pigs than type BB, difference is significant (P<0.01).5. By using SYBR Green Real-Time PCR to analysis different tissues of swine. All testing tissues show positive result. Expression of small intestine,muscle and cartilage are higher than other tissues.
