Three combinations (Huaixiang chicken♂×Huaixiang chicken♀, Huaixiang chicken♂×Princess chicken♀and Princess chicken♂×Princess chicken♀) were established in this experiment.The experiment was conducted to study the difference of the growth performance among the crossed combinations and their parents and to analyze single nucleotide polymorphism of extracellular fatty acid binding protein gene. Also, the associations between the detected SNPs and the growth performance were analyzed. One hundred and twenty chickens (male and female in half) were selected from every combination at birth and were randomly divided into four groups in every gender. The obtained results in the study were showed as follows:1. Comparasion of growth performance among three groupsThere were significant difference in body weight and feed conversion of chicken ( 16 weeks, in the same gender) among the three groups (P<0.05). Body weight in the three groups followed decreasing order as Huaixiang chicken (♂1744g /♀1590g), the hybrid chickens (♂1493g /♀1242g) and Princess chicken (♂1087g/♀894g), The feed conversion as Huaixiang chicken (♂4.11/♀4.00), the hybrid chickens (♂4.37g /♀4.07) and Princess chicken (♂4.65 /♀4.58).2. Comparasion of carcass performance among three groupsThere were significant difference in carcass weight, abdominal fatty rat and intermuscular fatty width among the three groups (P<0.05). Carcass weight in the three groups followed decreasing order as Huaixiang chicken, the hybrid chickens and Princess chicken. Abdominal fatty rate as Huaixiang chicken (5.90%), crossed chicken (5.22%), Princess Chicken (1.44%); intermuscular fatty width and subcutaneous fat thickness , respectively as Huaixiang chicken (1.30cm, 0.92 cm), crossed chicken (1.06cm, 0.57cm), Princess Chicken (0.73cm, 0.78cm).3. Comparasion of meat qulity among three groupsThere were significant difference in breast intramuscular fat and protein content between Huaixiang chicken and princess chicken (P <0.05), and followed as Huaixiang chicken (3.59%/88.78%) and princess chicken (2.83%/91.83%). There were not significant difference in other meat quality traits(P> 0.05).Intramuscular fat of females'breast muscle was significantly higher than cocks between sexes in the same combination, but protein content was significantly lower than males'(P <0.05). 4. Single nucleotide polymorphism analysis on chicken EX-FABP geneFive exons and the proximal flanking regions of the Ex-FABP gene were detected for SNPs with single strand conformation polymorphism method in 360 individuals from the three study groups. Sequence analysis indicated that thirty five novel single-nucleotide polymorphisms(GenBank accession nos. HM061617-HM061636) were found in chicken Ex-FABP gene. Among which three were in the 5'UTR, seven were in the coding region, and twenty-five were in the introns. Two SNPs in the coding region were missense mutant, and the other five were synonymous mutation.5. Associations analysis between the detected SNPs and growth performanceAssociations between fat traits and the SNPs were analyzed with general linear model, In locus T43C of exon1, chickens with genotype TC had higher values than that of genotype TT in hen population for intermuscular fat width (P <0.05). In locus G1472T of exon4, indivuals with genotype GG and TT had higher values than that of genotype GT in cock population for intermuscular fat width (P <0.05). The Two SNPs could be served as the candidate genetic markers in breeding. In locus C1985T of exon5, indivuals with genotype CC had higher values than that of genotype TT in hen population for subcutaneous fat thickness,intermuscular fat width and intramuscular fat(P <0.05). This site could serve as the candidate genetic markers in breeding.There were not significant associations between the fat traits and other SNPs.
