Puccinellia tenuiflora is a saline-alkali, drought-tolerant plant that is dense clumps perennial grasses, not only has the high feeding value, but also has the value of the construction of artificial turf and ideal grasses of biological treatment of saline-alkali soil.For investigating the resistance to Cd2+ stress, this study analyzed the SOD, POD and MDA of Puccinellia tenuiflora treated with Cd2+, The results showed that:the activity of the SOD and POD of leaves was improved under Cd2+ stress, and the accumulation of MDA was increased.Two suppression subtractive hybridization cDNA library of Puccinellia tenuiflora treated with Cd+ were constructed, and 33 differentially expressed gene segments from the forward and reverse cDNA library were obtained. Among them 26 segments were induced, and 7 was inhibited by Cd2+. A homologous analysis revealed that 29 gene segments had homology to the genebank, of which 25 had homology to the known transcripts, and 4 to unclassified proteins, the other 4 had no homology and might be new sequences.In the known function sequenses, 19 segments were induced and 6 was inhibited by Cd2+, of which such as GSR gene, y-ECS gene, membrane related protein CP5 gene and ABC1 family protein gene are relevant to the resistance of Puccinellia tenuifdora to Cd2+.Based on the sequences of ubiquitin-conjugating enzyme E2 gene, full-length cDNA of that was cloned from Puccinellia tenuiflora by using RACE method. Sequence analysis shows that the gene is 572bp in length, including 483bp of open reading frame, and encodes a protein of 161 amino acids, and 5'leader sequence, and 27bp 3'non-coding region. The bioinformations of ubiquitin-conjugating enzyme E2 gene also were analyzed and discussed.
