Telomere associated sequence (TAS) have a characteristic of species-specificity and can be used as genetic marker in chromosomal ends. Nine primers were used to amplify TAS from Glycine max, G. gracilis and G. soja. Special PCR products were amplified steadily by seven primers. Seven TASs, i.e., STAS-1452, STAS-1246, STAS-853, STAS-716, STAS-574, STAS-403 and STAS-379, were cloned and sequenced successfully. The results of bioinformatic analysis indicated that the sequences STAS-1452, STAS-853, STAS-574, STAS-403 and STAS-379 had characteristics of TAS. Blastn analysis demonstrated that they had high similarities between other soybean TASs in GenBank, but low similarities between TASs from other species. There were different copies of Arabidopsis-type telomeric repeat motif and many telomeric repeat motifs with mutations, as well as tandem repeats were found in these TASs. The STAS-716 and STAS-1246 were no similarities between other soybean TASs in GenBank, respectively. The STAS-716 had one Arabidopsis-type telomeric repeat motif, while no Arabidopsis-type motif in STAS-1246, but there were no the mutation motifs and the motifs in other species in the two sequences. Tendem repeat did not occur for the two TASs. Chromosomal localization analysis indicated that the TAS, STAS-574, shared by the three types of soybean was located to the proximal end of chromosome 3 in the Williams 82 soybean genome. Part fragments of STAS-1452, STAS-853 STAS-403, and STAS-379 were also located on the proximal ends of certain soybean chromosomes. STAS-716 and STAS-1246 were located in the interstitial, near-interstial, and distal regions of different chromosomes in the Williams 82 genome.
