Liver X receptorα(LXRα)is a class of lipid metabolism with the nuclear receptor, is an important lipid sensor.Its main function is through its corresponding receptor ligands of peroxisome proliferator and retinoic acid X receptors form heterodimers.Then the activation of the transcription of target genes under the control in a specific activator (oxidation steroid) ,thus affecting the body's lipid and carbohydrate metabolism, and by regulating the synthesis of fatty acids to regulate lipid homeostasis. Hepatocyte nuclear factor-4α(HNF-4α) is a hepatocyte nuclear factor (hepatocyte nuclearfactor, HNF) family member, highly expressed in liver, the family genes regulating the differentiation of liver cells play a key role, it also participates in liver glucose and lipid metabolism in the regulation of specific gene expression. LXRαgene and HNF-4αgene involved in regulating many aspects of life and metabolic activity in animals play an important role in growth and development.In this study, Yorkshire,Landrace,Wild boar,Yorkshire x Min pig,Beijin black pig large as the experimental material.Using PCR, cloned pigs LXRαgene and HNF-4αgene full-length coding sequence and intron 1,2,3,4,5,7,8 LXRα; Using the method of bioinformatics analysis LXRαgene and HNF-4αgene structure, function and molecular evolution domain; using PCR-SSCP method to analyze the LXRαgene and HNF-4αgene single nucleotide polymorphisms and to explore the different alleles .The distribution of the pig and its relationship with birth weight, weaning weight, birth length, birth body height, chest circumference at birth, muscle fiber diameter, muscle fiber density and muscle fiber cross-sectional area of the correlation analysis. The major findings are as follows:1.Boar LXRαgene complete coding region 1344bp, encoding 447 amino acids. The structure of nuclear receptor DNA exists binding and ligand binding. LXRαgene evolution more conservative, pigs and cattle homology was the highest (96.87%),and duck the lowest homology (70.47%).2.Boar HNF-4αgene complete coding region was 1425bp, encoding 474 amino acids. The structure of nuclear receptor DNA exists binding and ligand binding. HNF-4αevolution of a more conservative, pig and human homology of the highest (97.68%), and zebra fish, the lowest homology (81.01%)3.LXRαgene intron 1,2,3,4,5,7,8 were 646bp, 689bp, 748bp, 211bp, 304bp, 680bp4.LXRαgene in intron 2 and exon 4 were to find 5 SNP and one 2bp insertion / deletion, including exon 4 in the two single-core synonymous mutations are nucleotide mutation. Population genetics analysis showed that the detection of three sites in five varieties genotypes there were significant differences (P<0.01).5.Using PCR-SSCP method to find 5 SNP at intron 2, exon 8, intron 8 and exon 9 in HNF-4αgene , C→T at base 101978 in exon 8 is synonymous mutation, G→A at base 102001 in exon 8 is missense mutation, single nucleotide mutation T→C silent mutations in exon 9.Population genetic analysis showed that there were significantly different in the distribution of genotypes in 5 five breeds.6.LXRαgene genotype of TB3 with birth chest was significantly associated (P<0.05). LXRαgene genotype of TB4 with weaning weight and birth length was significantly correlated (P<0.05). HNF-4αgene genotype of SH1 with birth length was significantly related (P<0.01). HNF-4αgene genotype of SH2 with birth weight was significantly correlated (P <0.05).7.LXRαgene genotypes of TB2, TB3 and TB4 were not significantly relate with dmuscle fiber diameter, density and cross-sectional area (P<0.05). HNF-4αgene genotype of SH2 with muscle fiber diameter and muscle fiber cross-sectional area was significantly correlated (P<0.05).
