To observe the regulation of Nociceptin/Orphanin FQ (N/OFQ) and opioid receptor-like1 genes (ORL1 ) in energy metabolism ,this thesis selected mice epididymal adipose tissue as objection .We study the expession of N/OFQ and ORL1 genes in adipose tissue ,the levels of N/OFQ and ORL1 genes express in adipose tissue during different fasting time(24 hours and 36 hours) , and effect of insulin and daxamethasone on exprssion of N/OFQ and ORL1 genes levels .The results as follow:â… The levels of N/OFQ and ORL1 mRNA in adipose tissue in mice .The result showed: After amplification by RT-PCR ,the adipose tissue produced specific bands which were located 364bp and 563bp according to expectation .The result indicated that N/OFQ and ORL1 genes were expressed in adipose tissue in mice .â…¡Effects of fasting 24 hours and 36 hours on N/OFQ and ORL1 genes expression levels in adipose tissue in mice. The result showed: Compared with control group , expression of N/OFQ and ORL1 genes in adipose tissue significantly increased after fasting 24h (P<0.05) .Compared with fasting, expression of N/OFQ and ORL1 genes in adipose tissue decreased after refeeding 24h ,but not back to the control group. 2 ,Compared with control group , expression of N/OFQ and ORL1 genes in adipose tissue increased after fasting 36h ,but not significantly .â…¢Effects of hormone on N/OFQ and ORL1 genes expression levels in adipose tissue in mice .1, Compared to fasting 24h group, N/OFQ and ORL1 gene expression was low after insulin treatment, but not significant. 2, injection of dexamethasone lowered N / OFQ and ORL1 gene expression significantly (P <0.05) compared with the normal group.
