Rapid progress in genome sequencing projects provides information for the potential understanding of gene function, so effective strategies of founding gene function are increasingly in demand. So far many strategies have made Success. But the most important of them is building the mutant library. The mutant library is the basic material of functional genomics. China with a vast territory and complex clime have infinite wheat local variety, some organization has constructed wheat mutant libraries with local variety, but huanghuai region hasn't reported as the largest wheat yield zone. Furthermore, bread wheat has huge genome and more repeat sequence, so the wheat mutant library hasn't reached saturation in the world especially in our country at present. Therefore, constructing a new wheat mutant library with high yield and vast accommodate variety would accelerate wheat genome study in a certain extent, which could be used in wheat functional genomic research and provides useful information for wheat improvement. In this study, seeds of bread wheat Yunong 201 was treated by chemical mutagen Ethyl methane sulfonate (EMS). Biological characters and agronomic traits were investigated in M2 generation. Furthermore, several mutant individuals with deletion of different HMW-GS (High Molecular Weight Glutenin Subunit) were screened by SDS-PAGE. The results are as follows:1. Results indicated that 722 mutant individuals in total were obtained with the comparison of the wild type of common wheat cultivar Yunong 201. Phenotype mutants such as seedling habits, leaf appearance, plant height, spike appearance and mature period were obtained.The mutation frequency was 11.43%.2. Results indicated that a total of 139 leaf mutants were obtained, correspondingly mutant percentages was 2.20%.There are albino plant, small or big yellow spots on leaf, yellow leaf on the bottom, stripe on leaf, Verticillium and so on.3. Results indicated that a total of 220 stem mutants were obtained, correspondingly mutant percentages was 3.49%. There are Shorter mutants, higher mutants, stem thicker mutants, stem slimmer mutants, single tiller mutants, fewer tillers and more tillers mutants, clump dwarf mutants, half groveled mutants and so on.4. Results indicated that a total of 38 spike and grain mutants were obtained, correspondingly mutant percentages was 0.60%. There are short spike mutant, compact spikelet mutant, curve spike mutant, curve awn mutant, short awn mutant, big grain mutant, red grain mutant and so on.5. Results indicated that a total of 325 physiological character mutants were obtained, correspondingly mutant percentages was 5.15%. There are sterility mutant, maturity mutant, premature senility mutant, lethal mutant and so on.6. Results indicated that a total of 21 high molecular weight glutenin subunit gene mutants were obtained, correspondingly mutant percentages was 2.26%. There are mutants of A1, Dx5, Bx7 and Dy10.
