Objective: Study antitumor effect of novobiocin in liver cancer, and its modulation on clinical antitumor agents. Methods: The inhibiting effects of novobiocin and five anti- tumor agents alone and cotreatment in rat liver cancer cell line (H22) and human liver cancer cells were determined by antitumor agent sensitive assay (MTT method). At the same time we injected H22 cells to rats to establish transplanting tumors, then treated with novobioein, MMC and 5-FU for 3 weeks to investigate the rate of cell apoptosis and the tumor size using Tunel assay and tumor weighting. Results: The antitumor agent sensitive assay showed that antitumor effect of novobiocin was same to the five antitumor agents which was dose depandant. Novobiocin inhibition rate in H22 cell line was higher than those in MMC and CTX (PO.05), whereas in high dose novobiocin group was higher than MMC with significant difference (P
